ObjectiveBMT-1 belongs to benzimidazole bicyclic compounds,containing benzene and imidazole key groups.A large number of studies have shown that BMT-1 plays an important role in inhibiting the proliferation of various tumor cell lines,it can achieve anti-tumor effect by inducing tumor cell apoptosis.Until now,there is no report on acute B lymphoblastic leukemia treated by BMT-1.Therefore,this study aims to explore the effect of BMT-1 on proliferation,apoptosis and mitochondrial membrane potential of B lymphoblast cells,and to provide scientific and theoretical basis for the development and application of clinical acute lymphoblastic leukemia drugs.MethodsThe cell lines we used in our experiment were Daudi and Nalm6,and they were treated with different concentrations of BMT-1.(1)CCK8 assay was used to detect the effect of drugs on the cell proliferation ability.(2)The morphology change of Daudi was observed under an inverted fluorescence microscopy after the treatment by BMT-1(3)Apoptosis kit(including PI,Annexin V)was used to detect the role of BMT-1 on the apoptosis of Daudi;(4)The mitochondrial membrane potential was observed by Flow Cytometry(5)Western Blot was used to detect the expression of PARP and its related proteins.Results(1)The proliferation of Daudi and Nalm6 cells were inhibited by BMT-1 in a dose-dependent manner;(2)The cell density of Daudi cells treated with BMT-1 were significantly lower than those in the control group,this further confirm the cytotoxicity of BMT-1;(3)BMT-1 can clearly induce apoptosis of Daudi cells;(4)BMT-1 can clearly cause changes in mitochondrial membrane potential in a dose-dependent manner,this indicates that BMT-1 can induce the apoptosis of Daudi via mitochondria membrane potential pathway;(5)BMT-1 can cause Daudi cells to produce PARP cutters,the presence of this further suggests that Daudi cells do have apoptosis caused by BMT-1.ConclusionBMT-1 can effectively inhibit the proliferation of B lymphoblast cells,it can achieve this inhibitory activity by inducing apoptosis;The following observation of cell density change further suggested this.BMT-1 can also induce changes of mitochondrial membrane potential and the changes of PARP-related protein levels.
