ObjectiveIn this work,the airway was first infiltrated by inflammatory cells in patients with allergic asthma,the bronchoalveolar lavage fluid(BALF)was then collected and the total number of cells was counted and classified.Th1,Th2 and Th17 cells of peripheral blood mononuclear cells(PBMC)were detected by flow cytometry,and IL-10 and IL-17 levels of peripheral blood as well as IL-10 m RNA and IL-17 m RNA levels in mononuclear cells were determined via Enzyme-Linked Immuno Sorbent Assay(ELISA)and Fluorescence quantitative PCR method.Analyze the change of IL-10 and IL-17 when the airway was infiltrated by inflammatory cells,to provide the basis for the diagnosis and treatment of allergic asthma.MethodsSubjects were divided into two groups: the asthma group and the control group.The asthmatic group was responsible for 30 patients of allergic bronchial asthma without allergic treatment.After collection and centrifugation of the BALF in the asthmatic group,the cells were counted and the white blood cells were classified.The levels of IL-10 and IL-17 in the supernatant of the peripheral blood were measured by ELISA.At the same time,the peripheral blood mononuclear cells(PBMC)was used to determine IL-10 m RNA and IL-17 m RNA by Fluorescence quantitative PCR.Th1,Th2 and Th17 cell subsets in peripheral blood mononuclear cells were analyzed by flow cytometry.The same method was used for the study of the control group composed of 30 normal healthy people for comparison.Results 1.The number of cells in BALF and the percentage of lymphocytes and eosinophils in the asthmatic group were remarkably higher than that in the control group.The differences have statiscal significance.2.The result of Ig E in BALF of the asthmatic group was significantly higher than those in control group.The difference has statiscal significance.3.Serum IL-17 levels in asthmatic group were markedly higher than those in control group,while serum IL-10 in asthmatic group decreased compared with control group.The levels of IL-17 m RNA in peripheral blood mononuclear cells of asthmatic group were significantly higher than those in control group,but the levels of IL-10 m RNA in peripheral blood mononuclear cells of asthmatic group decreased compared with the control group,and the results were statistically significant.4.CD4+IL-4+ cells and CD4+IL-17A+cells in peripheral blood of asthmatic group were significantly higher than those in control group,and CD4+IFN-?+ cell level was significantly lower than that of control group.Conclusions When the airway was infiltrated by inflammatory cells for patients with allergic asthma,inflammatory cells in BALF and Ig E levels increased,Th1 cells in the peripheral blood decreased,and Th2 cells and Th17 cells increased.In asthmatic group,the levels of Serum IL-17 and IL-17 m RNA in peripheral blood mononuclear cells significantly rose,while the levels of Serum IL-10 and IL-10 m RNA in peripheral blood mononuclear cells decreased compared with control group,and the results with statistical significance.
