Expression Of Nupr1 In Gastric Carcinoma And Its Clinical Significance

Background:The incidence of gastric cancer is high in the world,because of its high transfer,fast proliferation and other characteristics,prompting rapid progress and poor prognosis of gastric cancer.The key for treatment of gastric cancer is early diagnosis,can significantly improve the survival rate of patients.But because of gastroscope and other invasive examinations are of high risk and high technology and equipment requirements,they are not easy to be widely accepted,abdominal CT and other noninvasive examinations have low early positive-rate.In the treatment,despite increasingly innovative surgical procedures and chemotherapy drugs and rapid development of targeted drugs,the gastric cancer patients get good effect,it still can not significantly improve survival prognosis of patients with gastric cancer,so there is an urgent need for a new diagnosis and treatment of gastric cancer.Due to the change of the microenvironment,such as hypoxia,injury,serum starvation,the body will produce the corresponding cellular stress response,so as to adapt to the environment to ensure the normal growth and function of cells.When the tissue cells are stimulated by stress,they will change the normal state of gene expression in response to emergencies,these genes playing the role in the stress response is called stress genes,cellular stress response requires the involvement of relevant stress molecules.In 1997,Nuprl was discovered by Mallo GV et al in rats with acute pancreatitis,it is detected high expression in pancreatic acinar,after sequencing,they got a 8kD sized molecule,named P8,which has a typical nuclear localization signal(NLS).It is also known as nuclear protein 1(nuclear protein 1,Nupr1).Nuprl is a kind of stress related nuclear molecule which has been shown a variety of biological functions,including the outstanding role in tumor development,it can play the role in transcriptional activity,showed high expression in pancreatic cancer,breast cancer,and related to tumor growth and metastasis.However,there are few studies about Nuprl in gastric cancer,and its expression in gastric cancer and its mechanism of action are not clear.Objective:Nuprl is a tumor related stress molecules.To detect the expression of Nuprl mRNA and protein in gastric cancer and adjacent tissues,to explore the relationship between Nuprl and the clinical significance of gastric cancer,provide new ideas for diagnosis,prognosis evaluation and effective treatment of gastric cancer.Methods:Real-time PCR and Western-blot was used to detect the expression of Nuprl,then we analyzed the relationship with the clinical characteristics of gastric cancer.Results:1.Melting curve drawing shows that fragment specific amplification,no amplification curve in negative control group can be discharged pollution factors.Real time fluorescent quantitative PCR results showed that the expression of Nuprl mRNA in 72 cases of gastric carcinoma(the relative expression was 2.0953±0.5997)was significantly higher than that in adjacent tissues,and the difference was statistically significant(P<0.05).2.The relative expression level of Nuprl protein in gastric cancer tissues was 2.0571±0.5426,which was significantly higher than that in adjacent normal tissues,and the difference was statistically significant(P<0.05).The expression of Nuprl protein in gastric cancer had consistent changing trends with mRNA expression.3.The differential expression of Nuprl in gastric carcinoma was correlated with tumor size,depth of invasion,lymph node metastasis,distant metastasis,degree of differentiation,pathological type and TNM staging(P<0.05),no significantly associated with sex and age of patients,the tumor site(P>0.05).Conclusion:Nuprl is highly expressed in gastric carcinoma,it is associated with the occurrence,invasion and metastasis of gastric cancer,Nuprl can be used as a new marker to evaluate the prognosis of gastric cancer patients,and it may become a potential therapeutic target for gastric cancer intervention.
