Purpose:this topic in human pancreatic cancer cells PANC-1 as the research object,were compared at different time,different concentrations of Jiawei Shengjiangsan on the inhibition of cell growth and proliferation in vitro,on cell apoptosis,while comparing the effect of curcumin and its main components.Through the preliminary study on the signal pathway and the molecular mechanism of pancreatic cancer cells treated by modified powder,to provide the theoretical and experimental basis for the clinical application of modified powder for the treatment of pancreatic cancer.Method:1 MTT was used to detect different concentrations of Jiawei Shengjiangpulvis(10,20,30,40,50mg/ml)and its main effective components of 24h,48h,72h after curcumin effect on human pancreatic cancer cell growth and proliferation of PANC-1,and compares the two inhibitory rate.2 using Annexin V-FITC and propidium iodide(promide iodine,PI)double staining method,the effect on cell apoptosis by flow cytometry were used to detect different concentrations of Jiawei Shengjiangpulvis and its main effective components of the effects of curcumin on human pancreatic carcinoma PANC-1 cells after 48 hours.3 study on using Western Blot method with different concentrations of Jiawei Shengjiangpulvis function of human pancreatic cancer cells PANC-1,P85,P110,AKT of P-AKT,the protein expression of PI3/AKT signaling pathway,and to investigate the molecular mechanism of Jiawei Shengjiangsan on human pancreatic cancer PANC-1 cells.Result:1 The Jiawei Shengjiangpulvis inhibitory effect on the proliferation of human pancreatic cancer PANC-1 cells to a certain extent in a dose and time-dependent manner,the low concentration group(10mg/ml)have no obvious effect,the high concentration group has obvious tumor cell proliferation inhibition effect,its efffect is better than that of the positive group and the main effective components of curcumin group results with statistical difference(P<0.05).2 The apoptosis rate of PANC-1 cells increased gradually with the increase of the concentration of 48h and the effect of curcumin on human pancreatic cancer cells.In the low concentration group,the apoptosis was not obvious,but under the action of the high concentration drug group,the cell morphology was changed,and the apoptosis rate was dose dependent.The apoptosis rate of human pancreatic cancer cell line PANC-1 was higher than that of the same group.3 For treatment of human pancreatic cancer cell PANC-1 by Jiawei Shengjiangpulvis after different concentration,the expression level of PI3K two protein subunits P85,P110 decreased with the increase of concentration,the total AKT expression level change trend is not obvious,and the phosphorylation of AKT especially in the high concentration group under(40mg/ml)expression levels were significantly lower.Conclusion:the modified powder can inhibit the growth and proliferation of human pancreatic cancer cell line PANC-1,and induce the apoptosis of human pancreatic cancer cell line PANC-1.The mechanism may be achieved by inhibiting the activity of PI3K/AKT signaling pathway,which has become one of the anti-tumor mechanisms.
