Nano hybrid materials have attracted extensive attention in the biomedical field.However,it is still a great challenge to fabricate a hybrid scaffold with homogenous distribution of biofunctional nanoparticles.In this research,histidine(His)was covalently cross-linked into collagen(Coll)scaffolds by 1-ethyl-3-(3-dimethyl aminopropyl)carbodiimide(EDC)/N-hydroxysuccinimide(NHS)to construct homogeneous Coll/His scaffolds where imidazole groups served as a template to chelate silver ions which were then reduced in situ to form silver nanoparticle(Ag NP)–hybridized Coll scaffolds(Coll/His/Ag NPs hybrid scaffold).Ag NPs uniformly embedded-hybrid scaffolds were characterized by SEM and XPS.TEM and DLS results revealed a uniform distribution of the Ag NPs(average size 7.2 nm)in the scaffold.The mechanical strengths of the scaffold were shown to increase with the introduction of Ag NPs.Moreover,the hybrid scaffold exhibited excellent biocompatibility-it could promote the growth and propagation of L929 cells,meanwhile maintaining robust antibacterial activities against both Gram negative and Gram positive bacteria.The Coll/His/Ag NPs hybrid scaffolds exhibited better anti-inflammatory properties and could promote dermal regeneration after being transplanted into the full-thickness infected wounds of SD rats.In this work,we offer a facile and simple method to fabricate the Coll/His/Ag NPs hybrid scaffolds for efficient healing of infected burned wounds in vivo.
