Objective:The normal microorganism and microorganism,microorganism and its host form a unified whole.Growth and development,metabolism and aging is closely related to microorganism.The ear canal is a small microecosystem,which is worth to be studied.This subject based on the exploring of the microbial community of ear canal is used to objective synergistic effect of the microbial community,to guide the use of antibiotics to cure Clinical ear infection.Methods:1.We choose the samples from both of the Department of Otolaryngology head and neck surgery of the First Affiliated Hospital of Dalian,from 2015 to 2016.The 29 cases are divided into two groups,13 patients in deafness group and 16 persons in normal group.2.We use traditional culture method and PCR-DGGE method to detect the biomass and diversity of the microflora in the ear canal of the normal elderly and the elderly with hearing loss.3.We use the automatic microbial identification and drug sensitive system to detect the drug sensitivity test of 2 groups.4.The results show that the number of CFU in 2 groups is the same.Deafness group is 71.5±10.73CFU/g,normal control group is 72.83±6.24CFU/g,there is no significant difference(P>0.05)Results:1.The results show that there is no significant difference between aerobic and anaerobic bacterial colonies in the 2 groups,which is 77.67±8.50 CFU/g,65.33±10.7 CFU/g,71.67±3.06 CFU/g,and 74.00±9.17 CFU/g.Two groups comparison,there is no statistically significant difference(P >0.05).2.After 24 h to 48 h,we find the apperance of bacterial colony is neat,white,with no hemolytic ring in Deafness group aerobic culture;dvisible circular convex,smooth surface,light yellow,a transparent hemolysis ring of colonies and white colonies around,no hemolysis ring in deafness group anaerobic culture;aneat,smooth,moist,white colonies in normal control group aerobic culture;smooth,moist,white colonies in normal control group anaerobic culture.3.We find the total amount of nucleic acid DNA is different.The results show that in the QIAamp DNA Stool Mini kit extraction,spectrophotometry,calculation,visible deafness group total nucleic acid DNA was 281.5±26.60(mg/m L),normal control group total nucleic acid DNA was 366.50±28.62(mg/m L),there is significant difference(P< 0.05),hearing loss group is lower than the normal group.4.From the results of PCR-DGGE electrophoresis and cluster analysis of microflora of the ear canal,we find the two groups are the same.The number of dominant bands in Deafness group is 28.46±2.76,and the normal group is 29.88±2.96,showing no differences.But the shading degree is different.5.By using Quantity One UPGMA dendrogram clustering softwarey,we find that microecological bacteria cluste of the deafness group and the control group of ear bacteria samples tested by PCR-DGGE maps cluster as a class.6.The results show that there are Staphylococcus epidermidis,Staphylococcus aureus,Corynebacterium diphtheria,Staphylococcus aureus,Streptococcus dentisani argensis,Propionibacterium acnes and Thermus tengchongensis in the ear canal of the aged.7.The results show that : Enterococcus faecalis is sensitive to penicillin,ampicillin,of aminoglycoside antibiotics resistant and susceptible to vancomycin;Staphylococcus aureus(methicillin resistant strains of MRSA bacteria to penicillin,gentamicin,)erythromycin,tetracycline,clindamycin,rifamycin resistant to vancomycin,nitrofurantoin and koalas for Ming sensitive Staphylococcus epidermidis;penicillin,erythromycin,chemitrim,vancomycin and linezolid sensitivity;Streptococcus pneumoniae to erythromycin,azithromycin,clindamycin,tetracycline,cotrimoxazole resistance that is sensitive to vancomycin and linezolid.Conclusion:1.There are no significant changes in the aerobic and anaerobic species of the microflora in the ear canal of the aged.The number of colonies in the deafgroup and the normal control group is the same.The DNA of deafness group is lower than that in normal control group.2.The diversity of microbial floras in two groups is the same.3.There are no special drug resistant strains of bacterial in the ear canal of the elderly.
