Aims: Gastric carcinoma(GC) is the second most frequently diagnosed cancer and the second leading cause of death in China. The tumorigenesis and progression of GC is a complex process caused by external and internal factors resulting in deregulation of the gene networks. Cytoplasmic polyadenylation element binding protein (CPEB) are sequence-specific RNA-binding proteins and the key factors controlling the elongation of the poly(A) tail and polyadenylation-induced translation. CPEB4 has been reported in association with tumor growth, vascularization and invasion of several malignancies.However, the related research is rarely reported on GC. Therefore, the characterization of CPEB4 in GC as well as the relationship between its expression level and clinical parameters will be important for developing novel cancer diagnostic and therapeutic strategies.Methods: In this study, Real-time PCR, immunohistochemistry and Western blot were applied to obtain the mRNA and protein levels of CPEB4 in GC and paired adjacent non-tumor tissues from 30 patients. Statistical analysis were performed with the Chi-square and Wilcoxon test.Results: CPEB4 mRNA was up-regulated to 90% in GC compared to the matched normal tissues. The expression level is correlated with lymph nodes metastasis,distant metastasis and pathological staging, but not related to age, gender,tumor location, tumor differentiation and tumor invasion. CPEB4 protein expression in GC was significantly higher than that in adjacent non-cancerous tissues. The expression level is correlated with lymph nodes metastasis, distant metastasis and pathological staging, and positively correlated with CPEB4 mRNA expression.Conclusion: CPEB4 mRNA and CPEB4 are increased in GC tissues relative to corresponding non-tumor tissues, and correlated with lymph nodes metastasis,distant metastasis and pathological staging. Our results indicated that CPEB4 acts as a carcinogen in the tumorigenesis and progression of GC.
