Background: There are a variety of hypotheses about the mechanism of "aging",which is more recognized free radical hypothesis. This hypothesis that active oxygen/nitrogen free radicals(ROS, reactive oxygen/nitrogen species) sustained role in the body of oxidative damage is the main reason leading to decreased physiological function.Glutathione, superoxide dismutase and other antioxidants play a role in scavenging free radicals under physiological conditions.The GSH/GSSG couple is regarded as the primary arbiter of the tissue redox state. SOD is the only antioxidant that specifically cleanses O2-free radicals. However, it is not the ability of the body to remove free radicals is stronger and healthier, which is due to the production and synthesis of many important substances need to participate in the oxygen from the base to carry out. Analysis of the normal range of GSH, SOD and other compounds helps to evaluate the redox status of the organism.Oral health conditions are often associated with general health status, such as diabetes and other diseases. Saliva is a safe, noninvasive and easily accessible body fluid.Objective: To study the derivatization conditions of GSH in NEM. Detection the age-related changes of GSH, GSSG and GSH/GSSG in saliva and whole blood, as well as the correlation between saliva and whole blood. To analyze the effect of age- and gender-related on superoxide dismutase inhibitory rate of saliva and plasma in healthy population and to study the relationship between the inhibitory rate of superoxide dismutase of saliva and plasma. In order to evaluate the overall health status of human by studying the above compounds in saliva.Methods: Healthy Chinese adults were inwestigated from the Department of Clinical Laboratory of Health Management Institute of General Hospital of Chinese PLA who were divided into young group(aged 20-44), middle age group(aged 45-59), elders group(aged 60-79), and grouped by gender. To optimize the derivatization conditions of glutathione and NEM in whole blood. Quantitative analysis of GSH and GSSG in saliva and whole blood was performed by liquid chromatography-tandem mass spectrometry(LC-MS/MS).We applied WST to detect SOD inhibitory rate in saliva and plasma.Results: The concentrations of GSH-NEM and GSSH in the saliva were 2.00±0.80?mol/L and 3.30±0.73 ?mol/L, respectively, and the ratio of GSH-NEM/GSSG was 0.63±0.27. The concentrations of GSH-NEM and GSSH in whole blood were 1493.41±243.95 ?mol/L, 0.79=10.34 ?mol/L, and the ratio of GSH-NEM/GSSG was 2312.90±1130.30. Experiments showed that GSH-NEM in saliva and whole blood were negatively correlated with age(r=-0.43 and r=-0.49, allP<0.05). Saliva and whole blood GSSG were positively correlated with age(r=0.32 and r=0.46, all P<0.05). It was noteworthy that the ratio of GSH-NEM/GSSG in saliva and whole blood was significantly negatively correlated with age(r=-0.56 and r=-0.53, all P<0.05). Meanwhile, there was a positive correlation between saliva and blood GSH-NEM, GSSG and GSH-NEM/GSSG ratio(r=0.28, r=0.28, and r=0.28 all P<0.05). Superoxide dismutase inhibitory rate in plasma and saliva in healthy adults were 61.27±5.79% and 43.40±12.99% respectively.Superoxide dismutase inhibitory rate in saliva(r=-0.49, P<0.05)and plasma(r=0.43,P<0.05 )has a negative correlation(P<0.05). There was a positive correlation between superoxide dismutase inhibitory rate in saliva and plasma(r=0.43, P<0.05). However the influence of gender was undetected(P>0.05).Conclusions: When the derivatization reagent NEM and the precipitant reagent SSA are added separately, a more thorough derivatization reaction can be achieved without additional adjustment of pH and temperature. Studies have shown that the GSH,GSSG,especially the ratio of GSH/GSSG in whole blood and saliva and the SOD inhibitory rate in plasma and saliva show age-related changes, and there was correlation between the above compounds in whole blood and saliva and plasma and saliva. Thus,there was a feasible way to estimate the oxidative stress status of body through measurement of GSH and SOD inhibitory rate in the saliva.
