1. The expression profile of miRNAs in tissue of pancreatic cancer.Aims: To explore the specific expressed and pancreatic cancer related miRNAs in the tissue of pancreatic cancer.Methods: Discovered the expression profile of miRNA in pancreatic cancer tissue by Affymetrix miRNAs microarrays to find the the different miRNAs among the two groups (pancreatic cancer tissues and adjacent tissues). All the targeted gene regulated by these miRNAs were acquired and analyzed by KOBAS system for target gene Disease, Pathway and GO enrichment analysis.Result: 10 tissue (5 cancer tissues and 5adjacent tissues) were carried out miRNAs microarrays analysis and screened 8 significant up-regulated miRNAs(fold-change > 10) and 1 down-regulated miRNA. Then we performed Disease, GO and Pathway Analysis which showed that target genes regulated by these selected miRNAs significantly associated with cancer signaling pathways, such as Pathways in cancer, MAPK signaling pathway, cellular binding, Wnt signaling pathway and Pancreatic cancer and many other signaling pathways. These results ensured us that the selected miRNAs were pancreatic cancer associated.Conclusions: Differentially expressed and pancreatic cancer-specific miRNAs in tissues screened by microarray analysis could be obtained by trend analysis of gene expression and functional analysis.2. The expression level and clinical significance of miR-31 in pancreatic cancer tissuesAims: To verify the relative abundances of the miR-31 screened by microarray in pancreatic cancer tissues and explore the correlations between the expression quantity of miR-31 and clinical features.Methods: Investigating the expression of miR-31 by real-time PCR in 12 pancreatic cancer and 9 adjacent matched tissues, These tissues had been collected from pancreatic cancer resection and all were diagnosed by General PLA Hospital during 2016.10?2017.3. The patients' clinicopathologic features (age, sex, smoking history, CEA, Cal 9-9, tumor location, tumor size, differentiated degree, lymph nodes metastasis) were collected. The relationship between the expression level of miR-31 and the clinical features of pancreatic cancer patients was analyzed.Results: The expression level of miR-31 was significantly higher in pancreatic cancer tissues than in adjacent tissues (p<0.05). There was no correlation between the abundance of miR-31 in pancreatic cancer tissues with the patients' gender,age,smoking history, tumor size, tumor location and the level of serum tumor biomarker(CEA and Cal 9-9). The expression level of miR-31 in cancer tissues was significant positive correlated with tumor differentiated degree (p<0.01) and lymph nodes metastasis (p<0.05).Conclusions:1, miR-31 expressed in pancreatic cancer tissue was higher than adjacent pancreatic tissue, which suggested the miR-31 acted as an oncogenic factor.2, High expression of miR-31 was closely related to lymph nodes metastasis and differentiated degree of pancreatic cancer, suggesting miR-31 played role in the process of the occurrence and development of pancreatic.
