With the changes in the people diet and life styles,obesity has become a serious worldwide epidemic.As obesity causes diabetes,hypertension,coronary heart disease and other dieseases,the World Health Organization(WHO)considers obesity and cancer as the most serious diseases to human health in the 21st century.Therefore,the research of anti-obesity has become a major topic of modern medical science.It was recently well recognized that intestinal microbiota was tightly linked with the health status of vertebrates.Researches indicated that food and drugs influence the intestinal mcrobiota as well,or even by changing intestinal mcrobiota to get influence on the body,especially obesity.However,whether the change of external environment can also change the intestinal microbiota,i.e.the internal environment,or through the change of intestinal microbiota to influence the metabolism and health status,is largely unknown yet.Our previous studies indicated that tea fusion fumigation showed distinct anti-obesity effects,tested with high-fat-diet induced obesity rats.The results imply that the external environment changed by tea fusion fumigation might also change internal environment,i.e.intestinal microbiota,as well.In this project,rats are fed with high-fat diet for establishing animal model of obesity.Fumigation temperature is set as(39±1)0C.Using the Pu-erh ripe tea.The tea fumigation(6.67%,w/w)were applied for 45-day.Genomic DNA was extracted from samples by DNA stool mini kit and Illumina high-throughput sequencing technology was used to sequence V6 variable region of the 16SrRNA gene.Combined with body weight index,serum TC,TG,HDL-C,LDL-C,FBS levels and ALT,AST activity,etc,of rats to explore the differences of gut microbiota diversity in Pu'er ripe tea fumigation process.The results showed that fumigation of Pu-erh ripe tea was able to reduce body weight Lee's index,perirenal fat pad weight,epididymal fat pad weight,fat weight,fat body ratio,serum TC,TG,LDL-C,FBS levels and ALT,AST activity,increasing HDL-C content.The results of liver sections showed fumigation of Pu-erh ripe tea can protect and refresh the liver damaged by high-fat diet.Compared with other stages the intestinal microflora diversity of obesity model construction stage was richer.When the rats into the weight loss stage,in the process of weight loss in the rat intestinal flora has changed,some of the high abundance of bacteria in the modeling stage began to decrease,while some of the low abundance of bacteria in the modeling stage began to rise.The Phylum Firmicutes was increased however bacteroides decreased in the tea fumigation.However,after the end of the tea fumigation experiment,the abundance of the Phylum Firmicutes was reduced.This project also opens a novel way to consume excessive tea products,and to prevent and treat obesity.
