Ginsenoside Rbl is one of main components from Panax ginseng, which has been reported to enhance neurogenesis and inhibit the neruoinflammatory cascade. Panax ginseng has been widely prescripted to treat affective disorders in China and other countries. The study was designed to determine the antidepressant-like effects of Ginsenoside Rbl in mice exposed to chronic unpredictable mild stress (CUMS), a model of major depressive disorder (MDD). After 4 weeks of CUMS, the mice exhibited depression-like behavior, then treated with Ginsenoside Rbl (10mg/kg,intragastric administration) for another successive 4 weeks. The antidepressant effect of Ginsenoside Rb1 was evaluated by sucrose preference test, the forced swimming test and tail suspension test. The Quantitative real-time PCR was used to determine the level of pro-/anti-inflammatory cytokines in cortex and hippocampus. The Immunohistochemistry was applied to detect the number of neuron (BrdU+/DCX+) in hippocampus. It was showed Ginsenoside Rbl reversed anhedonia in sucrose preference test and decreased immobility time in the FST,TST in CUMS-treated C57BL/6J mice. A dysregulation between pro-inflammatory cytokines (IL-1?, TNF-?and IL-6) and anti-inflammatory cytokines (TGF-?,Arg1 and IL-10) were detected in mice model of depression. After Ginsenoside Rbl intragastric administration,pro-inflammatory cytokines decreased and anti-inflammatory cytokines increased in cortex and hippocampus in MDD model mice. The numbers of newborn neuron in hippocampus were significantly increased in Ginsenoside Rbl-treated mice compared with saline mice. These data indicated that Ginsenoside Rbl had antidepressant-like effects, duing to it inhibiting inflammatory response and enhancing the hippocampal neurogenesis.
