Objective:To investigate the inhibitory effect of gallic acid on human adenovirus type four and its mechanism in vitroMethods:This study adopts a series of experimental methods.Cell culture and cytopathic effect observation assay were applied to determine the safe dose range of gallic acid,SPSS software probit regression was used to obtain the maximum non-toxic concentration(TC0)and the median toxic concentration(TC50);TCID50 assay were carried to determin the virulence of the adenovirus.The inhibitory effect of gallic acid on adenovirus in vitro was evaluated by the MTT cytotoxicity assay;Real-Time PCR and Western blot method were also used to analyze the inhibitory effect of gallic acid on HAdV-4's DNA and hexon protein respectively.Results:The maximum non-toxic concentration of gallic acid on A549 cells was TC0=32.75?M,the Median toxic concentration was TC50=279.906?M.The TCID50 of HAdV-4 in A549 cells was 10-6.5/100?L calculated by Reed-Munch method.The inhibition rates of four kinds of drug adding methods on adenovirus by MTT method were:the inactivation(73.19%*)>the adsorption(59.68%*)>the biosynthesis(13.06%*)>the cell protect function(3.15%")((*P<0.05),(#P>0.05)).The inhibitory effect measured by Real time PCR and Western blot on adenovirus is basically the same as that of MTTConclusion:The mechanism of gallic acid against human adenovirus type four is mainly depends its inactivation of virus in vitro thus affecting the adsorption effect of adenovirus on cell,which also had an inhibitory effect on human adenovirus type four's biosynthesis stage,mainly lies in the inhibitory effect on the synthesis of nucleic acid and protein of adenovirus,but it had no obvious effect on protect cells and prevent adenovirus invasion.
