Camellia oil is extracted from the seeds of Camellia oleifera(Camellia oleifera Abel)by means of pressing or leaching.Fujian is one of the main producing areas of camellia oil in China.Modern pharmacological research shows that camellia oil has the function of regulating immune function,improving blood circulation,anti-fatigue,antioxidation,blood lipids,blood sugar,anti-radiation,anti-bacterial and anti-inflammatory,preventing obesity,anti-tumor,and so on,the development of nutritional health function product of camellia oil has been paid more and more attention.This study is to study and compare the quality of camellia oil with different preparation technology and different producing areas,and to study the quality standard and stability of camellia oil soft capsules,in order to provide the experimental basis for quality control.I.Study on the quality of Camellia oilThis study was based on the national standard of the different preparation process of Camellia oil refractive index,relative density,saponification value,iodine value,acid value,unsaponification content and other physical and chemical indicators of determination and analysis;A HPLC method for determination of the main functional constituents of squalene in Camellia oil was established.The content of Squalene in Camellia oil was compared with different preparation techniques,and the composition and content of fatty acids in Camellia oil were compared by 26 kinds of control products accompanied by GC,and the HPLC fingerprint spectrum of Camellia oil from different habitats was established.The results show that:1.The acid number and peroxide value of Camellia oil in different preparation process were significantly different,acid value and peroxide value from high to low are leaching camellia oil,cold pressed camellia oil and hot pressed Camellia.2.Determination of the content of squalene in Camellia oil by HPLC,and Squalene has a good relationship within 1.0?200.0mg/L concentration range,precision,repeatability,stability rsd are less than 2%,the average recovery rate is 98.12%,The content of squalene in camellia oil was 0.0139%?0.0045%,and there is a significant difference,hot squeeze camellia oil in the squalene content is the lowest,only to dip in the oil of about 30%.3.GC determination results showed that the main fatty acid components of camellia oleifera oil and cold pressed camellia Oleifera were oleic acid,palmitic acid,linoleic acid,stearic acid,and peanut,which could detect ?-linolenic acid and linolenic acid from the Camellia oleifera,while the cold pressed camellia oil was not detected.4.10 batches of Camellia oil with HPLC fingerprint showed 9 common characteristic peaks,and the fingerprints of 5 batches of Camellia oil from Fujian were basically the same,and the overall similarity was higher than 90%,suggesting that the chemical constituents were close to each other.?.Study on the quality of Camellia oil compound soft CapsulesCamellia oil compound soft capsule is a kind of antifatigue functional food made up of camellia oil and glossy ganoderma spore oil at a certain proportion in the early stage of the study group.This study was to determine the refractive index,relative density,saponification value,iodine value,the content of no saponification and the chemical and physical properties of 4 batches of Camellia oil compound soft capsules.A HPLC method for determining the content of the main functional constituents in the compound soft capsules of Camellia oil was established,and the content of squalene in 4 batches of Camellia Oleifera compound was determined and the stability of camellia oil compound soft capsule was investigated for 3 months.The results show that:1.The mean acid number,peroxide value,saponification value,iodine value and unsaponification content of 4 batches of Camellia oil compound soft capsules were 2.11mg/g,5.31mol/kg,173mg/g,70g/100g and 20g/kg.2.HPLC method was used to determine the content of squalene in the compound soft capsules of Camellia oil,and squalene has a good relationship within 1.0?200.Omg/L concentration range,and the precision,repeatability and stability rsd were less than 2%,the average recovery was 93.86%,and the content of squalene in 4 batches of camellia oil compound soft capsules was 0.0067%?0.0071%3.On this basis,the quality standard draft of Camellia oil compound soft capsules was preliminarily formulated.4.Camellia oil compound soft capsule placed at room temperature for 3 months,its appearance,physical and chemical properties,squalene content and other indicators are not significantly changed,soft capsule stability is good.?.Study on antioxidant function of camellia oil compound soft capsuleCamellia oil compound soft capsule is developed as anti fatigue functional food,and the literature reports that Camellia Oleifera and Ganoderma lucidum spores have antioxidant effects.In this study,the DPPH free radical scavenging ability of camellia oil compound soft capsules was preliminarily tested,the results showed that camellia oil compound soft capsule had certain antioxidant function,and the IC50 of DPPH free radicals was 33.2mg/ml,which was equivalent to that of 0.03mg/ml VC scavenging DPPH radical.And with the increase of concentration,the effect of camellia oil compound soft capsule on the scavenging of DPPH free radicals increased gradually.The preliminary test results provide the basis for further development of the new antioxidant function of camellia oil compound soft capsules.?.Conclusion1.At present,the camellia oil is lack of activity or index components quantitative standard,this study established by HPLC method for the determination of squalene in Camellia oil,fast,simple and accurate,can provide the detection means for the quality control of camellia oils and related pharmaceutical products,and provide the basis for improving quality standard of Camellia oil.2.Camellia oil Preparation process of diagonal squalene,gamma-linolenic acid,linolenic acid and other active components have a significant impact,leaching camellia oil contains more active ingredients than the squeezing method of camellia oil more abundant,research and development of Camellia oil pharmaceutical products should be preferred leaching camellia oil.3.The HPLC fingerprint of Camellia oil in Fujian province is basically the same,with high similarity,its chemical composition,species and quantity are in general approximate,which provides scientific basis for the exploitation and utilization of camellia oil resources and the extension of the industrial chain in Fujian province.4.Camellia oil compound soft capsule quality is stable,controllable,good storage stability.5.The preliminary test shows that Camellia oil compound soft capsule has a better scavenging effect on DPPH free radicals,and provides experimental basis for further development of the new function of Camellia oil compound soft capsule.
