Objective: Effect of Toll Receptor 9(Toll-like receptors-9,TLR-9)on Acute Lung Injury Induced by Chest Trauma in Rats.Methods: Selected in April2016 to May purchased from Guizhou Medical University Experimental Animal Center 40 healthy male SD(Sprague-Dawley,SD)rats 40 rats were used as subjects.Using a small bio-impact machine(provided by the third Military Medical University).In the 500 kpa under the impact of the rat chest,the formation of severe lung injury.According to the AIS-90 standard,the injured rats were graded.After the injury,the rats were exposed to frequent,deep breathing,more obvious than before,shortness of breath,lips,delayed,apathetic,loss of appetite and decreased activity.The above tips: metabolic respiration in rats who have appeared,the purpose is to adapt to oxygen supply.Therefore,the use of 500 kpa impact of a better simulation of acute lung contusion in rats.Were randomly divided into five groups of eight: normal control group;chest trauma induced ALI group simulation(500kpa)5,24,48,72 hours after the impact,in the femoral artery bleeding to death.Mouse serum and lung tissue were collected.Elisa method was used to detect the expression of TLR-9receptor in serum and alveolar lavage fluid.The expression of TLR-9 protein in lung tissue was detected by RT-PCR(Reverse transcription-PCR,RT-PCR.The expression of TLR-9 protein in lung tissue was detected by HE staining.9,IL-6 receptor mRNA expression levels;Western blot was used to detect the protein level of TLR-9 in lung tissue.Results: Pulmonary histopathological examination showed that pulmonary edema,pulmonary congestion,neutrophil infiltration and inflammatory exudation were more obvious in the thoracic trauma group than in the blank control group.Immunohistochemical method showed that the color of the thoracic trauma group was significantly brown and yellow under the light microscope.No white wasobserved in the blank control group,and the rats were stained with 48 hours after thoracic trauma.Group mouse lung tissue chest trauma wet / dry weight ratio: chest rats as compared with the control group rats trauma,chest trauma rat lung wet / dry weight ratio was significantly lower than the control group.ELISA: serum and bronchoalveolar lavage fluid in the TLR-9,IL-6 expression levels,chest trauma set higher,the difference was statistically significant(P <0.05).RT-PCR shows Press:lung tissue of chest trauma TLR-9 mRNA,IL-6 mRNA expression than the control group increased,the difference was statistically significant(P <0.05).Western blot shown Press: lung tissue of chest trauma TLR-9 protein levels increased compared to control group,the difference was statistically significant(P <0.05).Conclusion:The increased expression of TLR-9 after trauma in rats,suggesting that TLR-9 participates in the inflammatory response of pulmonary contusion.
