Objective:To investigate the expression of Nek8 mRNA and the expression of Nek8,BICD2 and VHL proteins in colorectal cancer tissues,and to evaluate relationship between these three proteins and the clinicopathological characteristics and prognosis of colorectal cancer.Methods:The expression of Nek8 mRNA in 30 pairs of colorectal cancer tissues,adjacent colorectal cancer mucosal tissues(3centimeters away from the tumor tissues)and adjacent normal colorectal mucosal tissues(8 or 10 centimeters away from the tumor tissues)were detected by RT-PCR and RT-qPCR methods.The immunohistochemical method was used to detect the expression and pathological features of Nek8,BICD2 and VHL proteins in 30 pairs of normal colorectal mucosal tissues and colorectal tumor tissues.At the same time,the expression of Nek8,BICD2 and VHL proteins were detected by immunohistochemistry in 20 cases of chronic inflammation of colorectal mucosal tissues,20 cases of colorectal tubular adenoma tissues(level II)and 60 cases of colorectal tubular adenocarcinoma tissues,which was observered the correlation with clinicopathological parameters.Results:(1)The expression of Nek8 mRNA in colorectal cancer tissues and adjacent colorectal cancer mucosal epithelium were higher than those in adjacent normal colorectal mucosal epithelium.The expression of Nek8 mRNA in colorectal cancer tissues was the highest in these three kinds of different pathological types.(2)With immunohistochemical results,there were high positive expression rates of Nek8 and BICD2 proteins in 30 pairs of colorectal cancer tissues[56.7%(17/30),76.7%(23/30)],which was higher rates than those in adjacent normal colorectal mucosal tissues[13.3%(4/30),13.3%(4/30)].The positive expression rate of VHL protein in colorectal carcinoma tissues[30.0%(9/30)] was significantly lower than that in adjacent normal colorectal mucosal tissues [63.3%(19/30)](P<0.05).(3)The positive expression rates of Nek8 and BICD2 proteins in colorectal adenoma tissues [80.0%(16/20),75.0%(15/20)] and colorectal adenocarcinoma tissues[83.3%(50/60),73.3%(44/60)] were obviously higher than those in chronic inflammation tissues [25.0%(5/20),20.0%(4/20)].There existed a downward trend about the expression of VHL protein in these three kinds of different colorectal pathological types of colorectal adenocarcinoma tissues,colorectal adenoma tissues and chronic inflammation tissues [75.0%(15/20),40.0%(8/20),26.7%(16/60)](P<0.05).There were no relationship between these three proteins and age,sex,infiltration degree,lymph node metastasis,clinical stage,five year survival rate and metastasis(P>0.05).(4)Comparing in pairs of Nek8,BICD2 and VHL proteins in colorectal cancer tissues,there existed the significant correlation between Nek8 and BICD2(P < 0.05),which was positive,and the other pairs did not exist statistical significance(P>0.05).Conclution: The expression of Nek8 and BICD2 are increased in colorectal cancer tissues.The expression of VHL is decreased in colorectal cancer tissues.Nek8,BICD2 and VHL may act the role in tumorigenesis and development of colorectal cancer.
