In this paper,we used cotton thread serve as the substrate to fabricate cotton thread immunochromatographic assay device for lung cancer biomarkers——ferritin,squamous cell carcinoma antigen(SCCA)and carcinoembryonic antigen(CEA)detection.In order to study of cotton thread immunochromatougraphic assay device,Gold nanorode(GNR)and gold nanoparticles-enriched carbon nanotube nanocomposite(CNT/GNPs)were modified with different detection antibody forming reporter probe.The main points of this thesis are summarized as follow:(1)Electrochemical detection of human ferritin based on gold nanorod reporter probe and cotton thread immunoassay deviceIn this study,a natural cotton thread immunochromatographic assay device combined with gold nanorod(GNR)reporter probe is designed for electrochemical detection of human ferritin,a lung cancer related biomarker.Human ferritin was used as an analyte.The capture antibody(cAb)is applied to the test zone and the detection antibody(dAb)is used for the preparation of GNR-dAb.An enhancement of the sensitivity is achieved by using gold nanorod as an electroactive report probe compared with a traditional gold nanoparticle(GNP)report probe.Under optimal conditions,the device is capable of measuring 1.58 ng/mL ferritin which meet the requirement for clinical diagnosis.(2)Novel gold nanoparticles-enriched carbon nanotube nanocomposite probe based immunochromatographic assay on cotton threadIn this paper,we present the first application of gold nanoparticles decorated carbon nanotube(CNT/GNPs)nanocomposite reporter probe in an immunochromatographic assay on cotton thread.SCCA and CEA are used as an analyte,monoclonal detection antibody is used for the preparation of the corresponding CNT/GNPs nanocomposite reporter probe,monoclonal capture antibody bound tightly to the surface of cotton thread.The low-cost,sensitive and rapid detection of SCCA and CEA can be achieved by immune reaction between antibody and antigen.Quantitative detection are based on the reading of colored bands with scanner and"Image J" software.Results indicate that the sensitivity of the method was improved than traditional gold nanoparticles(GNPs)reporter probe.
