Objectives:In this study,under the guidance of traditional Chinese medicine and modern medicine on nonalcoholic steatohepatitis,from the inhibition of NF-? B signaling pathway activation,the activation of NF-?B signaling pathway of the relevant factors as the research points to Ik B-? Phosphorylation and subsequent ubiquitination mechanism and related influencing factors as the target,through the help of modern medical technology,at a deeper level to explore the role of electroacupuncture suppression NF-? B signaling pathway activation mechanism,observed by electro-acupuncture on high-fat diet And to investigate the effect of electroacupuncture on the expression of phosphorylated I?B-? protein in patients with nonalcoholic steatohepatitis,and to explore the effect of electroacupuncture on non-alcoholic steatohepatitis Mechanism and mechanism of electroacupuncture treatment of nonalcoholic steatohepatitis in order to provide a new experimental basis for the clinical application of electroacupuncture in the treatment of nonalcoholic steatohepatitis.Methods: Forty male Wistar male rats aged 8 weeks were randomlydivided into 2 groups: normal group(n = 10)and model group(n = 30).The normal group was fed with standard diet(70% carbohydrate,20% protein,10% fat).The model group was fed with high fat diet according to Fan Jiangao [1].The standard diet was based on 2% cholesterol,10 %lard).After 12 weeks,two rats were randomly selected from the normal group.Six rats were randomly selected from the model group.The fresh liver tissues were removed from the rats.The pathological changes of the liver tissues were observed under light microscope.After the liver pathology was confirmed,The rats in the remaining model group were randomly divided into model group,electroacupuncture group,drug group,8 rats in each group.Normal group,the model group to continue feeding before,without any treatment;electroacupuncture group to continue high-fat diet feeding,daily application of electro-acupuncture therapy to stimulate NASH rats Zusanli,Hong Leong,liver Yu,Taichong points;drug group continued high(UDCA)250mg / kg / d aqueous solution.All rats were anesthetized with 2% sodium pentobarbital overnight,and blood was collected from the inferior vena cava.The fresh liver tissues were harvested and the rats were sacrificed.The rats were sacrificed at room temperature,(ALT,AST),blood lipid(TG,TC,FFA)and rat liver tissue were measured by electroacupuncture and western medicine ursodeoxycholic acid on NASH rats.The liver histopathology,liver function Phosphorylation of I?B-? protein and pathological changes of liver tissue.Results:1.liver index: model group,electro acupuncture group and medication group was significantly higher than that of the normal group,P < 0.05);Electroacupuncture group than the model group decreased significantly,P < 0.05);no significant differencesbetween drug group and EA group(P > 0.05).2.liver function(ALT,AST): the model group was significantly higher than normal group(P < 0.05);Electroacupuncture group was significantly lower than the model group(P < 0.05);Electroacupuncture group than in drug group decreased,but no significant difference between the two(P > 0.05).3.blood lipids(TG,TC,FFA): the model group was significantly higher than the normal group(P < 0.05);the electric acupuncture group was significantly lower than the model group(P <0.05);there was no significant difference between the electroacupuncture group and the drug group(P > 0.05).The expression of liver tissue 4.phosphorylation of I kappa B-alpha: compared with normal group,model group of I phosphate kappa Balpha protein expression level increased(P < 0.05),use after EA treatment,phosphorylation of I kappa B-alpha protein expression levels were significantly reduced(P < 0.01);compared with the drug group and model group,phosphoric acid I kappa B-alpha protein expression levels also decreased significantly(P < 0.01),decreased as the EA group,but no significant difference between the acupuncture group and drug group(P>0.05).Conclusions:Electroacupuncture therapy and drug ursodeoxycholic acid treatment of non-alcoholic steatohepatitis rats were significantly improved symptoms;electro-acupuncture therapy for non-alcoholic steatohepatitis in rats than the use of drugs ursodeoxycholic acid,Electroacupuncture can significantly reduce the liver index,lower blood lipids,improve liver function.The level of phosphorylated I?B-? protein in the model group was up-regulated,and the expression level of phosphorylated I?B-? protein was significantly decreased after EA treatment,suggesting that the dissociation of I ? B-? oxidative phosphorylation increased in non-alcoholic steatohepatitis,The incidence of hepatitis,electroacupuncture can reduce the dissociation of I?B-? protein,inhibit the activation of NF-?B,which may be an important mechanism of electro-acupuncture treatment of non-alcoholic steatohepatitis.
