[Research background]: Nonalcoholic steatohepatitis(NASH)is a very important stage in the course of nonalcoholic fatty liver disease(NAFLD),which directly affects the development trend of the disease and the prognosis of the disease.However,the pathological mechanism and treatment of NSAH are not fully understood.[Research purposes]: The study is based on the experimental animal model of NASH rats induced by high-fat diet.To give those ill rats amount of saline or different concentrations of Yin Qin Qing Gan Decoction solution by gavage in the experimental process.We can observe the changes of living habits and physical characteristics of the rats which been gived the high fat diet and application of Yin Qin Qing Gan Decoction.In the experiment the rats were killed at the end of part to collect its liver tissue samples and blood samples.We measured the pathological changes of liver tissue and the levels of blood lipid,liver function and cytokine.Then based on specific data statistical analysisto evaluate the curative effect of Yin Qin Qing Gan Decoction on NASH rats.On signal transduction mechanism which marked by the release of the cause of interleukin-6(IL-6)and tumor necrosis factor-?(TNF-?)as the starting point to explore the molecular mechanism of Yin Qin Qing Gan Decoction in the treatment of NASH.Based on the investigation and analysis of the literature,this paper discusses the rule of TCM syndrome and treatment of NAFLD,the characteristics of pathogenesis and the pharmacological effects of the decoction.[Research operation]: Divided 40 healthy male SD rats were all about 250 g which purchase from the experimental animal center of Hubei province the weight into 4 groups randomly.Respectively named as blank control group(K group),model group(M group),low dose of Chinese medicine group(D group)and high dose of Chinese medicine group(group G)and give each experimental rat to mark markers to distinguis.The rats in the normal control group were fed with normal diet,while the other 3 groups were fed with high fat diet according to the standard dose within 1 weeks.The rats were started treating with intragastric administration after the establishment of NASH model induced by high-fat diet.The rats in the normal control group and the model group were given the right amount of normal saline,and the high and low dose groups of traditional Chinese medicine(referred to as the treatment group)were treated with different concentrations of Yin Qin Qing Gan Decoction.In experiment 2nd and 4th weeks of the weekend,randomly selected 5 rats from in each experimental group and killed one by one.Blood samples and liver tissue samples were taken from experimental rats.Then preserved separately and later to do the relevant inspection.Serum cholesterol(TC),the triglyceride(TG),aspartate transaminase(AST),Glutamic-pyruvic transaminase(ALT),Interleukin-6(IL-6) and Tumor necrosis factor-?(TNF-?)were detected and compared with.Statistical analysis is also important.[Research result]:(1)The weight of the rats which in the high dose and low dose Chinese medicine group and the model group increased.The fur was rough,and the body shape was significantly higher than that of the normal control group after 1 weeks of feeding with high fat diet.The fur was rougher,and the body shape was significantly higher.The rats were sacrificed and the liver tissue observed by naked eye visible throughout the liver color rendering of yellow soil and yellow spots,see section after the incision with yellowish greasy,liver lipid deposition,crisp texture,typical fatty degeneration.The rats were sacrificed and its liver tissue observed by naked eye visible throughout the liver color rendering of yellow soil and yellow spots,see section after the incision with yellowish greasy,liver lipid deposition,crisp texture,typical fatty degeneration.(2)The rats' which had been established the NAHS model by high-fat diet,its TC&TG lever is higher than the blank control group(P < 0.01).But,because of has been given the treatment of Yin Qin Qing Gan Decoction,its TC&TG lever is decreased(P<0.05).With the extension of treatment time,The serum TC and TG values of the rats in the traditional Chinese medicine group were significantly lower than those in the low dose group(P <0.05).(3)The AST and ALT of liver tissue in three groups fed with high fat diet were significantly higher than those in blank control group(P < 0.01).The levels of AST and ALT in the model group were significantly lower than those in the control group(P <0.05).With the prolongation of treatment time,the serum AST and ALT values of the two groups were lower than those of the low-dose Chinese medicine group(P <0.05).(4)The levels of TNF-? and IL-6 in the three groups fed with high-fat diet were significantly higher than those in the blank control group(P <0.01).The serum levels of TNF-? and IL-6 in the two groups of rats treated with Yin Qin Qing Gan Decoction were significantly lower than those in the control group(P <0.01).With the prolongation of treatment time,the serum levels of TNF-? and IL-6 in the two groups were significantly lower than those in the control group.[Research conclusion]:(1)The liver histological morphology of the three groups of rats fed with high fat diet was higher than that of the blank control group.The changes of lood lipids and liver function suggest that high fat diet may play a key role in the development of rat NASH.(2)The use of Yin Qin Qing Gan Decoction in the treatment of high-fat diet-induced NASH rats can reduce the serum TC,TG,improve liver function.Reduce the level of cytokines TNF-?,IL-6;improve the pathological status of liver tissue,suggesting that Yin Qin Qing Gan Decoction may be by reducing the release of cytokines,thereby protecting the liver from chronic liver injury and inflammation in NASH.
