Experimental Study Of "Yiqi Huoxuefa" In The Treatment Of Ruptured Lumbar Disc Herniation

Objective: 1.To investigate the effect of P38 MAPK phosphorylation on the expression of MMP-3?MMP-7 in rat model of ruptured lumbar disc.2.To observe the influences of Yiqi Huoxue decoction on intervertebral disc and P38 signal pathway in rat model of ruptured lumbar disc,and to explore its Preliminary molecular mechanism.Methods: Establish a rat model of lunber intervertebral disc.and then all rats were divided into model group,yiqi Huoxue Decoction group,Yiqi Huoxue+blocker group and blocker group,And a sham operation group?n=10?was set up.The sham operation group and model group were intragastrically given purified water.and other groups were given corresponding drugs for 3w.the rats were killed after 3w,The content of TXB₂ and PGF1? in serum and histopathological changes were detected by Enzyme-linked Immunosorbent Assay?ELISA?and HE staining.The expressions of P-p38 MAPK and MMP-3,MMP-7 in intervertebral disc were detected by Western-Blot.Results: 1.Pathologicol observations in rats after the modeling of nerve root compression of nerve tissue,see mild interstitial edema,a little more blood vessels,mononuclear inflammatory cell infiltration and a small amount of fibrous tissue proliferation,sorne of nerve atrophy.by Yiqi Huoxue decoction treatment,the lesion improved ratings have dropped.2.Model ratsby Yiqi Huoxue decoction therapy.Plasma TXB₂ levels and TXB₂/PGF_1?ratios were significantly decreased,while the PGF1 a level was elevated,showed a positive change.compared with model group,the P values were<0.01,significant difference in the result.3.Content of p-P38 MAPK and MMP-3,MMP-7 phosphorylation level increased in Yiqi Huoxue decoction group compared with model group?P<0.05 or P<0.01?.Content of p-P38 MAPK and MMP-3 ?MMP-7 phosphorylation level increased in Yiqi Huoxue decoction+ blocker group compared with blocker group?P<0.05 or P<0.01?.Conclusion?s?: By Yiqi Huoxue decoction treatment,the lesions improved ratings have dropped.which may be related to increasing phosphorylation levels of MMP-3 and MMP-7 by activation of P38 signal transduction pathway.
