Objective: To investigate the intervention mechanism of Anchang Decoction on metabolic markers of colonic mucosa in ulcerative colitis rats by metabolomics detection method based on meteorological and mass spectrometry.Methods: 1.using 2,4,6-trinitrobenzene sulfonic acid(TNBS)modeling,TNBS dose 90mg/kg,the establishment of ulcerative colitis rat model.2.The optimal model of ulcerative colitis in SD rats was randomly divided into model group,Lizhu enteric group,American salabazine group(positive drug control group),low decoction medium,high dose group(treatment group).3.The rats in each group were sacrificed at the5 th,10th,and 15 th day after treatment.The rats were sacrificed and the mucosal tissues of the colonic mucosa were taken for meta-chromatographic-mass spectrometry.The principal component analysis and partial least squares(OPLS-DA),and finally the VIP value(threshold>1)of the first principal component of the OPLS-DA model was used to find the differential metabolic rate(P<0.05)Things.Results: 1.The obtained chromatograms were matched with the mass spectrometer to identify 47 qualitative metabolic compounds.The multivariate statistical analysis showed that the second time point(day 10)was the most successful treatment of the ulcerative colitis model,Anchang Tang treatment group overall treatment effect is better than the positive drug control group,of which the effective dose of Anchang Tang in the best.2.After identification of the differences,the potential metabolic markers of colonic mucosa in rats with ulcerative colitis were obtained,including adenine,cytosine,uridine,glycine,glucose,xylose,β-alanine,nectar Sugar,lactic acid,sarcosine.3.There were 10 metabolites in the Anchang Tang treatment group and the callback rate was significantly higher than that of the positive drug group(P<0.05).The moderator of Zhongchang Tang was the best.Conclusion: 1.It is feasible to establish ulcerative colitis rat model with 2,4,6-trinitrobenzene sulfonic acid solution.2.The model is optimal for the second week after modeling,and the initial treatment time is available.3.There were 10 potential metabolite markers in colonic mucosa of rats with ulcerative colitis and normal rats: adenine,cytosine,uridine,glycine,glucose,xylose,β-alanine,mannose,Lactic acid,sarcosine.4.The effect of amlazapine on intestinal inflammation was better than that of Lizhu Changle group and Anchang decoction group.The effect of Lizhu Changle group on intestinal flora was better than thatof Meisharazine group and Anchang Tang treatment group.Anchang Tang middle dose group the best effect.5.Anchang Tang may regulate the intestinal environment by reducing the levels of adenine,cytosine,uridine,lactic acid,glycine and glucose,increasing mannose,xylose,sarcosine and β-alanine levels,Colitis against colitis,enhance cellular immunity and regulate the therapeutic effect of intestinal flora.
