Objective:In this study,we tried to disclose the effects of oncostatin M(OSM)on the activation and proliferation of hepatic stellate cell line T6(HSC-T6),and detecte the expression of oncostatin M receptor ?(OSMR?)in CDAA diet-induced nonalcoholic fatty liver disease(NAFLD)of rats,then explored its possible mechanism.Methods:In vitro,HSC-T6 cells were cultured for 1 day,4 days and 7 days respectively and then ?-smooth muscle activator(?-SMA)and OSMR? were detected using western blotting.HSC-T6 were cultured in 96 wells for 4 hours,then adding 0,5,10 and 20ng/ml of OSM respectively and cultrued for another 24 hours or 48 hours.At last,adding 10?l Counting Kit-8(CCK-8)reagent for 2 hours.The OD value was measured at the wave length of 450nm.In vivo,thirty male SD rats,6 weeks of age and weighing 160gą20g were obtained and fed a week to adapt to the environment.They were randomly divided into two groups with 15 rats in each group.The normal control group were fed with basic diet and the model group were fed with choline-deficient L-amino acid-defined(CDAA)diet.Twelve weeks later,the rats were anesthetized with ether and the right lobe of the liver was taken and used for the production of pathological specimens and the detection of the target protein.The expression of a-SMA was detected by immunohistochemical and OSMR? in liver tissue sections was checked by western blotting.Results:1.With the prolongation of HSC-T6 culture,the expression of ?-SMA and OSMRp in HSC-T6 increased(P<0.05).2.The addition of OSM could promote HSC-T6 proliferation(P<0.05),and its effect was dose and time dependent.3.The level of a-SMA in liver of group CDAA was higher than that in normal control group(n=15,P<0.05),and the expression of OSMR? in liver of CDAA group was higher than that of normal control group(n=15,P<0.05).Conclusions:1.As HSC-T6 activation,the expression of OSMRP increased.2.OSM could promoted HSC-T6 activation and proliferation.3.With CDAA induced hepatic fibrosis in rats,the experession of OSMR?increased in liver.OSM might act on HSCs through OSMR? to promote NAFLD devlopment.
