Objective:To study the the expression and clinical significance of Klotho in the serum and urine of IgA nephropathy patients.Methods:Twenty four patients who were diagnosed as primary IgAN by renal biopsy in Yanbian University Hospital from 2013 to 2016,including 15 males and 9 females,the average age was 49,13±15.76.Ten healthy people were used as the control group,including 6 males and 4 females,the average age was 44.80±13.61.Respectively,to detect the expression of Klotho by ELISA in healthy people and IgAN patients with serum and urine.And the laboratory data was collected,including systolic blood pressure,diastolic blood pressure,body mass index,total Cholesterol,triglyceride,low density lipoprotein cholesterol,high density lipoproteins cholesterol,blood urea nitrogen,Serum creatinine and uric acid.The datas were analyzed by SPSS23.0.The values were expressed as mean ± standard deviation.The differences between two groups were compared using the t test.To analyze the correlation between klotho protein and clinical datas by Pearson.Results:1.The levels of SBP,DBP,ALB,CHO,TG and LDL-C in IgA nephropathy group were significantly higher than those in control group(P<0.01).2.Serum and urine Klotho levels in the IgAN group were lower than those in the healthy control group(P<0.05).3.The level of serum Klotho in IgAN was negatively correlated with DBP(r =-0.471,P =0.020),CHO(r=-0.483,P = 0.017),LDL-C(r=-0.526,P=0.008).Conclusion:1.The level of Klotho in serum and urine of IgAN patients was lower than that of healthy controls,suggesting that the decrease in Klotho level was associated with the progression of the disease.Klotho may be involved in the progression of the disease by activating the expression of the RAS components,activating the inflammatory responses,damaging the glomerular endothelium,and promoting kidney fibrosis.2.There was a negative correlation between Klotho and DBP,CHO and LDL-C in serum of patients with IgAN,suggesting that the higher the level of Klotho in serum,the less the degree of hypertension and hyperlipidemia,and the mechanism may be that Klotho,by directly decomposing fat cells,Quality deposition,reduce vascular calcification,but also may directly stimulate the secretion of NO,diastolic blood vessels,thereby reducing the glomerular endothelial injury,delay the development of the disease.
