Effect Of Turmeric Volatile Oil On The Proliferation And Apoptosis Of Human Skin SCC A431 Cells

Objective: To investigate the effect of turmeric volatile oil on the proliferation of skin squamous cell carcinoma A431 cell and explore the mechanism of its apoptosis.Methods: Different concentration of turmeric volatile oil were applied on skin squamous cell carcinoma A431 cell;Detection of proliferation inhibition rate by CCK-8(Cell Counting Kit-8);Morphological changes of cells were observed by inverted microscope;Apoptosis was evaluated with AO/EB double staining and flow cytometry(FCM);Western blot method to detect Caspase-3,Caspase-9 protein expression..Results: To investigate the activities of turmeric volatile oil with different concentrations(0,5,10,20,40,80 umol / L)on human skin squamous cell carcinoma A431 cell after cultured for 24 h,36h,48 h By cell counting kits(CCK-8).It showed increasing inhibitory effect with drug concentration ranged from low to high in a dose dependent relationship.After treated for 48 h,when the drug concentration was up to 10 mg / L,Irregular cell morphology were presented under inverted microscope after treated with turmeric volatile oil,and the cell proliferation speed of A431 cells were declined as well.There was visibled part of the early apoptosis cells,nuclear chromatin yellow-green or yellow after AO/EB staining.When the drug concentration reached at 20 mg / L,there was karyopyknosis,part of the tumor cell membrane began to break down and part of the contents of cells were released;early apoptosis cells was visibled yellow fluorescence and late apoptosis cell orange was visibled fluorescent.When the drug concentration was up to 40 mg / L,the normal structure of the tumor cells was unable to see,some cells into pieces,and there is a lot of floating cells;a large number of nuclei chromatin dead cells and debris was orange after AO/EB staining.Flow cytometry instrument testing indicates that turmeric volatile oil on the skin squamous carcinoma A431 cells can promote apoptosis.And with the increase of drug concentration,the drug concentration-apoptosis rate is positive correlation?est results show that the expression of Caspase-3 and Caspase-9 with the drug concentration increasing,in a certain range within the scope of concentration dependence.And under the action of the same drug concentration,amount of Caspase 3 protein expression were more than Caspase-9.Conclusion: Turmeric volatile oil has an effect of proliferation inhibition and apoptosis-inducing on human skin squamous cell carcinoma A431 cell.The mechanism of apoptosis may be related to increasing the expression of Caspase 3 and Caspase-9.
