Background:DAVFs are pathologic anastomosis between dural arteries and cerebral venous,the sustained hypertension may lead to cerebral hemorrhage,cerebral edema or nerve dysfunction.Complex DAVFs is still one of the hardest diseases on clinic,and it's important to confirm the etiological agent.In the study of DAVF etiology,the angiogenesis caused by venous hypertension was found to play pivotal role.To control the process of DAVFs,it's key point to confirm the role of angiogenesis in the DAVF pathogenesis.Purpose:1.Dectect the shear stress change in model rats with different venous hypertension.2.Detect the expression of AIBP and VEGF in rat model with different venous hypertension and HUVECs cultivated under different shear stress.Explore the effect of shear stress changes resulting from venous hypertension on AIBP-regulated angiogenesis.Methods:First section: Establish rat model with different venous hypertension,80 rats were divided in to 4 groups.Group A was performed right carotid artery and external jugular vein anastomosis and occlusion of left transverse sinus vein,group B was performed right carotid artery and external jugular vein anastomosis,group C was performed ligation of right carotid artery and right external jugular vein,group D was sham group,all rats were performed the craniotomy of parietal bone to expose superior sagittal sinus.Measure and calculate the shear stress of superior sagittal sinus after 3 months,and execute rats to detect the expression of AIBP and VEGF.Second section: According to different shear stress,HUVECs were divided into 5 groups.Group A-E were cultivated under10,5,2.5,1,0 shear stress respectively for 24 hours.Then detect the expression of AIBP and VEGF in the cell supernate and cholesterin in cells.This experiment was repeated three times in each group.Results:First section: The shear stress of group A is 5.32±0.31dyn/cm2,group B is 2.39±0.025dyn/cm2,group C is 0.97±0.005dyn/cm2 and group D is 0.88±0.009dyn/cm2,the change of shear stress is caused by the flow victory in superior sagittal sinus.The results of PCR and Western show the AIBP in dural mater of group A and B decreased obviously,and the expression of VEGF increased.Compared with group D,there is significant difference.There is no significant difference between group C and group D.Immunohistochemical results shows that AIBP was expressed in the whole vascular layer.Second section: There is significant different between the expression of AIBP and VEGF in A,B and C group and group E,P<0.05.The expression of AIBP in A,B and C group is lower than group E,and the expression of VEGF in A,B and C group is higher than group E.There is no significant difference between group D and group E.Conclusion:1.The shear stress of superior sagittal sinus increased in venous hypertension rats model after 3 months,and the expression of AIBP in dural matter decreased after 3 months.2.The expression of AIBP of the HUVECs cultivated under high shear stress decreased and VEGF expression raised,and the cholesterin raised in cells.It points out that the shear stress may inhibit the expression of AIBP to activate VEGF up-regulation.
