Objective: Rheumatoid arthritis is a kind of chronic autoimmune diseases that affects human health and quality of life.In this study,we estimated the anti-inflammation,anti-analgesic and anti-rheumatoid arthritic effects of YC-5,as well as the primary underlie mechanisms.Methods:(1)The evaluation of the anti-inflammation effect of YC-5 in vivo: The anti-inflammatory activities of marine steroid compounds YC-5 were evaluated by rat ear-swollen induced by xylene test and cotton ball-induced granuloma formation test.(2)The evaluation of the anti-analgesic effect of YC-5 in vivo:The analgesic effects of marine steroid compound YC-5 were observed by thermal stimulation test and acetic acid-induced writhing response in rats.(3)The effects of YC-5 on CIA model: Through the collagen-induced arthritis model on rats,the arthritis score index of SD rats and the influence on histopathological changes of SD rats' ankle were studied to evaluated the effects of Marine steroid compounds YC-5.(4)The research on the primary anti-rheumatoid arthritic mechanisms of YC-5: The levels of serum TNF-?,IL-1?,PGE2,MDA and SOD were measured by ELISA and the NF-?B of synovial tissues were examined by immunofluorescence.Results:(1)Marine steroid compound YC-5 significantly inhibits the rat ear-swollen induced by xylene(F=8.65,P<0.01)and granuloma formation by cotton ball induced(F=30.31,P<0.01).(2)Marine steroid compound YC-5 significantly inhibits the pain induced by hot stimulation(F=55.37,P<0.01)and obviously prolong the writhing latent period(F=25.03,P<0.01)and reduces writhing times(F=23.84,P<0.01).(3)Marine steroid compound YC-5 could obviously reduces the arthritis index score(F=546.95,P<0.01),and significantly reduces synovial hyperplasia the collagen-induced arthritis in the ankle joints.(4)Marine steroid compound YC-5 significantly reduces the infiltration of inflammatory cells,and reduces NF-?B p65 expression in the pathological synovial tissues(F=76.77,P<0.01)and content of TNF-?(F=157.17,P<0.01),IL-1?(F=100.13,P<0.01),PGE2(F=164.48,P<0.01)in serum,as well as enhances the activation of the serum SOD(F=14.42,P<0.01),and lowers the content of MDA in serum(F=22.92,P<0.01).Conclusion: The study indicates that marine steroid compound YC-5 has an effective therapeutic action on rheumatoid arthritis,which may relate to the mechanism that marine steroid compound YC-5 decreases the release of inflammatory factors to inhibit oxidative stress and improve synovial inflammation by adjusting the NF-?B signaling network related inflammation factors.All above suggest that marine steroid compound YC-5 may be used as a potential anti-rheumatoid arthritis candidate drug.
