objective: To investigate the correlation of NCX1 calcium iron channel protein and mRNA,autophagy associated proteins and mRNA,and apoptosis pathway associated protains and mRNA in hyperlipidemic induced acute pancreatitis;and provide a new clue for pathogenesis of hyperlipidemic associated acute pancreatitis.Methods : 0 male healthy cleaning grade SD rats completely randomly devided into 4 groups:Hyperlipidemia induced acute pancreatitis group(GA,N=10),Non-hyperlipdemia acute pancreatitis group(GB,N=10)),Hyperlipidemia group(GC,N=10))and Blank control group(GD,N=10));each group involves 10 rats.12 hours later,After modeling hyperlipidemia with 10% Tyloxapol solution injected to caudal vein of mice in GA and GC,then,retrograte injection of 3.5% sodium taurocholate solution into biliopancreatic duct through duodenal wall establishing AP models on GA and GB rats,another 12 hours later,sampling blood and pancreatic tissues for later experiments.Blood samples are used to testing triglyceride(TG),cholesterol(TC),blood amylse(AMY),blood lipase(LPS);one part of pancreas used to evaluating tissue injury with hematoxylin and eosin stain(H&E stain),and another part of tissue used to testing NCX1?LC3??Beclin-1?Caspase-3 and Caspase-9 protains expression difference among 4 groups by use of western-blot assy;NCX1?LC3??Beclin-1?Caspase-3 and Caspase-9 mRNA expression are detected by Real-time quantitative Polymerase Chain Reaction(q RT-PCR).Results:(1)Models of hyperlipidemia are successfully made in GA and GC,TG and TC level are significantly higher than GB and GD,(P <0.05);but,TG and TC level in GA and GC is no more difference between them,so does in GB and GD.(2)AMY,LPS as inportant serological indicators of acute pancreatitis,they are much more increasing in GA and GB compared with GC and GD,(P < 0.05).as for GA and GB,there are no more difference between them,and the same for GC and GD.(3).Histopathological score are significantly higher in GB,as compared with GC and GD(P < 0.05);while GA are higher than GB(P < 0.05);GC and GD almost equal with each other.(4)Comparing with GC and GD,the protein and mRNA expression quantity of NCX1?LC3??Beclin-1?Caspase-3 and Caspase-9 in GA and GB are much more higher,(P < 0.05).But for the NCX1 expression,whether protein or mRNA GA and GB are non-differential;differen from NCX1 expression in GA,expression of LC3 II ? Beclin-1 ? Caspase-3 and Caspase-9 in GA are significantly up-regulated compared with GB,GC and GD for each other;the protein and mRNA expression quantity of NCX1 ? LC3 II ? Beclin-1 ? Caspase-3 and Caspase-9 in GC and GD are also no difference.Conclusion: NCX1 channel participate in acute pancreatitis,but not the crucial important one in hyperlipidemia-induced acute pancreatitis;autophagy involves in hyperlipidemia-induced acute pancreatitis according to LC3 II ? Beclin-1expression level.Caspase-3 and Caspase-9 are much higher in hyperlipidemia-induced acute pancreatitis reveal apoptosis and necrosis are more drastic in it.
