This study was designed to investigate the protective effects and possible mechanism of tetramethylpyrazine(TMP)isolated from Ligusticum chuanxiong on diabetic nephropathy in rats.First,removed the right kidney of all rats through operation.Then,Diabetes was induced in male Sprague-Dawley rats by a single intraperitoneal injection of 30mg/kg of streptozotocin.Sprague-Dawley(SD)rats were divided into six groups: contol,model,TMP15,TMP30,TMP60 and IRB80.Two weeks later,the dose of different TMP were administered intragastric gavage daily for 8 and 12 weeks.Examined the blood glucose,urine volume,body weight,water consumptition consumption and renal function of all rats.were examined in the begining,4th,8th,and 12 th week.Pathology morphological,LK/BW,immunohistochemistry and western blotting were determined in 8th and 12 th week.Treatment after 8 and 12 weeks.Compared with model group,blood sugar,urine output,water quantity were lower(p < 0.01)in TMP30?TMP60 and IRB80 group.High TG levels,and high TCHO hematic disease were significantly improved(p < 0.05)in TMP30?TMP60 and IRB80 group.BUN and Scr,24 h urine protein values were decreased(p < 0.05)in TMP30?TMP60 and IRB80 group,but the model and 8 weeks IRB80 showed no statistical significance.HE,PAS,Masson positive expression to reduce and relieve glomerular structure destruction in TMP30?TMP60 and IRB80 group.TGF-beta 1,alpha SMA and FN protein expression were significantly decline,protein semi-quantitative analysis has significant difference(P < 0.05)in TMP30?TMP60 and IRB80 group.and at the same time analysis of TGF-beta 1,alpha SMA and FN AOD value(r = 0.838,P < 0.0001 and r = 0.922),P < 0.0001,indicating that TGF-beta 1 was a positive correlation with alpha SMA and FN.Our results suggested that administration of tetramethylpyrazine(TMP)can reduce kidney damage caused by diabetes.
