Objective:In this study,the mice with adenosine A1R knockout gene were taken as the research object to observe the effect of intraperitoneal injection of caffeine on acupuncture analgesia and to explore the mechanism of the effect of caffeine on acupuncture analgesia.Methods:The mice were divided into four groups:normal group,model group and acute caffeine group,model plus caffeine group.Then according to the different genotypes of mice in four groups,each group was divided into wild mice group,heterozygous group and homozygous group which a total of twelve groups.Each group contained 15 mice with a total of 180 genetic mice.The normal group without any treatment;acute model in mice intraplantar formalin(20ul,1.5%),then mice plantar were swelled with white transparent vesicles.The mice stopped licking could be regarded as successful model.Caffeine group received intraperitoneal injection of caffeine(10mg/kg)modeling.Caffeine group was first modeled and then deal with intraperitoneal injection of caffeine.The large group of four taken the electrophysiological detection firstly and examined to observe the changes of acupuncture before and after acupuncture intervention 10min mechanical pain threshold and heat pain threshold.Then take the observation of acupuncture intervention before femoral head two muscles reflex and the EMG changes of the two biceps femoris in 0-5min after 1min intervention.Results:1.Behavior tests showed:In the control group and the formalin group,the analgesic effect of the hybrid and wild mice was more significant(p<0.01)than homozygote.In the caffeine group,formalin injecttion and caffeine injection group,the hybrid and homozygote had no statistical difference before and after acupuncture(p>0.05),however,mechanical and thermal hyperalgesia was observed in the wild mice in two groups when compared with the other kind of mice,having a significant statistical difference(p<0.01).Comparing four groups respectively,the therapeutic effect of acupuncture in both heterozygous and wild mice of the control group was better(p<0.01)than caffeine group,the formalin group was better(p<0.01)than formalin injection and caffeine injection group,homozygote mice had no significant statistical difference(p>0.05).2.Electrophysiological experimental results:In four groups,acupuncture could not produce analgesic effect on homozygotes(p>0.05)within 0-5 minutes in three different acupuncture intensity;the analgesic effect was more better on heterozygous within 0 minute(p<0.01)in the control group and the acupuncture manipulation was stronger than 1 TC and 0.5 TC.In formalin group,the effect of the 0-2min was best(P<0.001),however,3-5min effect was not obvious(P<0.05)and 0.5TC has the best intensity effection.Hyperalgesia appeared in the caffeine group(p<0.05)when compared with formalin injection and caffeine injection group(p>0.05).In the control group and the formalin group,the analgesic effect of 0-2min was the best(p<0.001)on the wild mice,and the analgesic effect in the formalin group was better than that in the control group(p<0.01).While in the caffeine group,formalin injection and caffeine injection group,there was no obvious analgesic effect but with hyperalgesia occasionally.The analgesic effect of the control group was superior(p<0.01)to which of the caffeine group.And the formalin group was superior(p<0.01)to which of the formalin injection and caffeine injection group.Conclusion:1.Adenosine A1 receptor was not only involved in the regulation of pain,but also played an important role in acupuncture analgesia.2.Caffeine antagonized adenosine and A1 receptor can weaken the acupuncture analgesia.3.Acupuncture analgesia was more sensitive to mechanical pain than thermal pain.
