Objective:To study the exogenous b FGF combined with modified dingzhi pill prescription on scleral remodel of Guinea pigs during the development of formed privation myopia influence.Method:(1)50 three weeks old healthy male guinea pig were divided into five parts:Blank control group as group A,group B is myopia model controlgroup,group C is modified dingzhi pill filling stomach group,group D is b FGF injected posterior scleral group,group E is b FGF combined modified dingzhi pill.(2)Except group A,other groups were given continuous privation translucent goggles left eye three weeks established privation myopia(FDM)model,in the time of the experiment fourth week,group C and D and Efor three weeks of drug intervention:which C group set records modifie dingzhi pill irrigation stomach 1ml/100 g,once a day,total 21 times;Group D posterior pole Tenon's injection b FGF,weekly once,total three times;Group E modified dingzhi pill irrigation stomach once a day,Posterior pole Tenon's injection b FGF,weekly once.(3)On the 1st,14 th,21th,28 th,35th days of the experiment,diopters and axis length were measured completed by a professional technician and recorded the data.When the experiment 22 nd days(The successful model on the first day),43 rd days(the last once measurement)used 10% chloral hydrate solution executed random sampled animals,took the posterior scleral,the choroid,and the retinal,preparationorganization pathology slice,respectively for HE dyeing and masson dyeing,compared FDM model before and after change and the drug intervention every group under optical microscope every layer thickness and the differences of the sclera collagen fiber in number and array.(4)Digimizer image analysis software measurement under light microscope,HE slices the centra-l area of scleral thickness.Results:(1)Form-deprivation myopia sclera after three weeks,scleral disorder of collagen fibers,the gap of fiber becomes larger,lamellar structures is not clear,partial or complete dissolution of collagen fibers,a thinner thickness.(2)Form-deprivation myopia sclera after six weeks,model group posterior sclera significantly thinner,choroid,and retinal slightly thinner,collagen fiber split and dissolve,the interstice between collagen fiber becomes larger,the number of collagen fiber is sparse,arranged disorder;The group of modified dingzhi pill sclera thickness no obviously change,collagen fiber mild disorder,visible part collagen clearance;b FGF injection group sclera and the choroid thickness no obviously changes,retinal particles layer clear,thickness normal,sclera collagen fiber mild disorder;The combined group each layer thickness no obviously changes,collagen fiber arranged neatly.(3)Compared with the blank group(104.4±2.31um),three weeks model group(99.14 ±1.83um)scleral thickness was significantly attenuated,there was a statistically significant difference(P<0.01);six weeks model group(93.47±2.22um)compared with three weeks model group,scleral thickness was significantly thinner,the difference was statistically significant(P<0.01);Both the group of modified dingzhi pill(97.97±2.29um)and b FGF injection group(98.44±1.92um)sclera were thicker than six weeks model group(93.47±2.22um),the difference was of significance in statistics science(P<0.01);The group of modified dingzhi pill and b FGF injection group with thickness,the difference is not obvious,no statistically significant difference(P>0.05).Combined group(101.36 ± 2.47um)thicker than group of modified dingzhi pill and b FGF injection in thickness,existed statistically significant difference(p<0.05).Conclusion:(1)Translucent patch method of blocking light to reach the retina,serious damage to animal form succeeded in establishing a form-deprivation myopia.(2)With the form-deprivation time extends scleral tissue morphology under the microscope with diopter and axis parallel change,suggesting that the change of diopter is mainly caused by the over extension of the axis of the eye,while the change of the axis of the eye is the result of the scleral remodeling.(3)Local inject exogenous b FGF combined with oral administration of modified dingzhi pill can effective control of sclera remodeling,inhibit or delay the occurrence and development of myopia-pig.
