| Objective:To investigate the correlation of SERCA2 a with myocardial injury,arrhythmia,syndrome differentiation of deficiency and excess by observing Xintongfang’s effect on expression of SERCA2 m RNA,myocardial histomorphology,arrhythmia and death rate in acute myocardial infarction rats.Methods: 40 SD rats were randomly divided into sham operation group,model group,Xintongfang group and Diltiazem control group.The myocardial infarction model was established by using ligation method.Expression of SERCA2 m RNA was detected by RT-PCR method,arrhythmia was tested by Electrocardiogram,morphological changes were observed by HE staining,syndrome differentiation of deficiency and excess was made by scoring criteria.Results: The expression of SERCA2 am RNA were obviously increased in Xintongfang group(P﹤ 0.05),the incidence of arrhythmia was decreased(P < 0.05),Significantly reduced myocardial damage.The degree of myocardial injury was negatively correlated with SERCA2 m RNA expression(r=-0.78,P=0.00);Expression of SERCA2 m RNA in group without arrhythmia was significantly increased(P<0.01),and expression of SERCA2 m RNA was positively correlated with syndrome differentiation score(r =0.969,P=0.00).Conclusion: Xintongfang has a protective effect on acute myocardial i nfarction in rats.The mechanism of the effect may be related to its in creasing of expression of SERCA2 am RNA in rat,which can help decre ase the incidence of arrhythmia;SERCA2a has protective effect on my ocardial tissue,and SERCA2 a expression is associated with arrhythmia and syndrome differentiation of deficiency and excess. |
