| Objective:To study the expression of CD200 in acute myeloid leukemia patients and the relationship of clinical efficacy and the white blood cell counts when they be diagnosed.Furthermore,to compare the levels of IL-17,IL-4,IFN-yin serum and the level of FOXP3/CD4+ T Cells in bone marrow before and after chemotherapy.Methods:The expression of CD200 on leukemic stem cells(leukemic stem cells,LSCs)in bone marrow were detected by flow cytometry in 51 cases newly diagnosed acute myeloid leukemia(acute myeloid leukemia AML)hospitalized in the first affiliated hospital,zhejiang university of medicine.Using the same methord the cells expressing FOXP3 +/CD4 + were detected before and after chemotherapy in patients,bone marrow.Besides,IL-17,IL-4 and IFN-γlevels of serum were also detected by enzyme-linked immunosorbent assay(ELISA)in the control group and in AML patients before and after chemotherapy.The relationship of the expression of CD200 on leukemic stem cells in acute myeloid leukemia and the prognosis related factors such as the high white blood cell count and the first complete remission rate.The change of the immune factors such as IL-17,IL-4 and IFN-ywere observed before and after chemotherapy as well as the cells expressing FOXP3 +/CD4 + in bone marrow.Results:Compared with the complete remission(CR)group,the expression of CD200 on leukemic stem cells of non-complete remission(NR)group when newly diagnosed was significantly higher(3.10±1.64vs4.76±2.65,p<0.05).At the same time a significantly higher expression of CD200 in high leukocyte count group than non-high leukocyte count group was observed(4.79 ± 2.83vs3.72 ± 2.23,p<0.05).IL-17,IFN-ylevels were significantly decreased before chemotherapy in acute myeloid leukemia patients compared with the control group(24.10 ± 8.55vs32.06 ± 9.09,p<0.05 147.05 ±42.29vs427.72 ± 67.84,p<0.05),and significantly increased after chemotherapy vs before(331.78 ±67.52vs427.72 ± 67.84,p<0.05;31.08 ± 11.72vs32.06 ± 9.09,p<0.05).At the same time the levels of IL-4 and FOXP3+/CD4 + cells in acute leukemia patients were significantly increased compared with the healthy people before chemotherapy(142.10 ± 54.41vs59.26 ± 28.77,p<0.05;7.48 ± 2.41vs4.18 ± 2.43,p<0.05),and decreased significantly after chemotherapy(83.79 ± 31.37vs142.10±54.41,p<0.05;5.22 ± 2.39vs7.48 ± 2.41,p<0.05).Conclusions:The expression of CD200 is related to poor prognosis factors such as poor effects of chemotherapy and high white blood cell counts.The immune-related factors as IL-17,IL-4,IFN-yand FOXP3/CD4+ in acute myeloid leukemia patients will significantly change before and after chemotherapys,suggesting that the immune of T lymphocytes play an important role in the anti-tumor mechanisms;and CD200+ affects the effect of therapy and the prognosis by inhibiting the cellular immune functions. |
