The Chinese yam widely exists in most areas in China, rich in resources, compendium of materia medica think that yam can "beneficial kidney, spleen and stomach, diarrhea, collapsed stripping, embellish fur". Yam by its unique medical value is widely used in traditional Chinese medicine. Also because of the delicious taste, it is a daily dining tables, the life of the food processing. Yam has many active components, such as polysaccharide, yam saponins, allantoin, Dehydroepiandrosterone, flavonoids, etc. Based on flavonoids in yam as the research object, to optimize the extraction technology of flavonoids in the yam yam are analyzed, the structure of flavonoids, and use of flavonoid compounds in yam do ?-glucosidase inhibition test in vitro, the main results were as follows:(1) in the process of yam flavonoids extraction, leaching time, liquid ratio and concentration of ethanol yam flavonoids to obtain all have great influence. Flavonoids mention gain rate increased with the increase of leaching time, peak and 60 min. Increase as the auxiliary extraction time, flavonoids obtained rate leveled off, a slight decline trend. With the increase of liquid ratio, flavonoids obtained rate sharply increase, to 10:1 peak, increase and liquid ratio, rate of flavonoids to have leveled off. Higher ethanol concentration, significantly increased the rate of extraction of flavonoids, around 60% reached the peak, and ethanol concentrations continue to rise, flavonoids obtained rate reduced emerged obviously.(2) Constructs the comprehensive score of yam flavonoids extraction in the regression equation:Y=1.85-0.006024X1+0.068X2+0.12X3+0.021X1X2+0.012X,X3-0.043X2X3-0.085X12-0.1X22-0.12X32 (R2=0.9850) Can be seen from the equation of the impact on the rate of flavonoids to mention made in yam is larger liquid ratio and auxiliary extraction time, the effect of ethanol concentration. To optimize the extraction process parameters as follows: 60% ethanol concentration, auxiliary extraction time 64 min, the liquid than earth (mL/g). The regression analysis results show that the regression model of the determination coefficient R2=0.9850, the model can be used for extracting flavonoids from the yam prediction control and the conditions of extracting result.(3) After purification, through the observation of flavonoid compounds in yam ultraviolet spectral characteristics that the structure of flavones and flavonols, add different diagnostic reagent, after observing the redshift, infer that there may be 4'-OH or 7-OH,4'-OH or 2-OH, there are 5,5,6,7-2 OH structure or 5 proposal or 3,3',4'-three OH, A and B ring on the adjacent two phenol-OH, there are 5-OH structure.(4) Through a-glucosidase inhibition test in vitro, can see the flavonoids in yam to a-glucosidase has certain inhibitory effect. Yam flavonoids in the concentration of 0.5 mg/mL-3 mg/mL of a-glucosidase inhibitory effect is better, and even slightly higher than acarbose. Back from 3 mg/mL, yam inhibition rate of flavone and a-glucosidase didn't have obvious concentration-response relationship, and acarbose and a-glucosidasee inhibition rate still present stable concentration-response relationship, the inhibition effect of total flavonoids of yam is lower than the inhibition effect of acarbose. Also can be observed in the yam flavonoids of a-glucosidase inhibition effect is better than that of different concentrations of ethanol elution of yam medium flavonoids components inhibitory effect.
