The objective of this article is to research the chemical constituents of Valeriana jatamansi Jones and neuroprotective effects,which is solid foundation for innovative drug discovery in neuroprotection based on treating neurologic disorder by regulating qi-flowing for strengthening spleen.It will greatly be significant for development of new drug to neurodegenerative disease.With percolate extraction,dry powder of Valeriana jatamansi Jones was extracted by 75%ethanol.Then the extract was extracted by petroleum ether,ethyl acetate and n-butanol.The ethyl acetate extract was separated preliminary with silica gel chromatographic methods.Then,various chromatographic methods such as silica gel,Sephadex LH-20,RP-C18 column chromatography,MCI column chromatography and RPLC was used over and over again to separate and purify the compounds.The structures of these compounds which were elucidated by HRESIMS,'H NMR,13C NMR,DEPT,2D NMR and compared with the known compounds.Twenty-five compounds were isolated from the ethyl acetate extract.Identified as 8-O-isovaleroxyl-1-O-?-D-glucopyranosyl jatamanin J(1),prinsepoil(2),8-hydroxypin oresinol(3),pinoresinol(4),2,5-methanocyclopenta-1,3-dioxin-7-ol(5),vibutinal(6),3-(4-hydroxy-3-methoxyphenyl)-propenal(7),baldrinal(8),11-ethoxyviburtinal(9),double-tetrahydrofuran lignans-4-O-?-D-Glucopyranoside(10),pinoresinol-4'-O-?-D-glucoside(11),(7S,8R)-dehydro coniferyl alcohol-8-5'-dehydroconiferyl aldehyde-4-O-?-D-glucopyranoside(12),4-hydroxybenzyl-?-D-glucopyranoside(13),methylarbutin(14),magnolol(15),din-butylphthalate(16),4-[(E)-3-ethoxyprop-l-enyl]-2-methoxy phenol(17),eugenyl-O--D-glunopyranoside(18),ethylconiferin(19),palmitic acid(20),protocatechuic acid(21),3-(4-?-D-glucopyranosyloxy-3,5-dimethoxy)-phenyI-2E-propenol(22).5-hydroxymethyl furfural(23),daucosterol(24)and 8-hydroxypinoresinol-4'-O-?-D-glucopyranoside(25).Among the total,iridoids including five,lignans including eight,phenylpropanoids including five,one sesquiterpene,two phenolic acids,one fatty acid and three categories compounds.Compound 1 is identified as new structure by searching data-base of SciFinder Scholar,It noteworthy that compound 1 is a new secoiridoid glycoside ring-opened between C-1 and C-2 produced by the cleavage of the pyran ring isolated from natural sources is relatively rare.Compounds 12,15,18 and 19 were isolated from Valerianaceae for the first time,Compounds 6,7 and 16 were isolated from V.jatamansi for the first time,Compound 23 was isolated from this plant for the first time.According to the traditional application and modern research of Valeriana jatamansi in nervous system.The compounds were tested in vitro neuroprotective activities against CoCl2 induced PC 12 cell damage model and the method of CCK-8 was used to evaluate its neuroprotective activities.The results showed that compounds 1,5,6,9 have potential neuroprotective effects,among them compound 1,5,6 can significantly improve the PC 12 cells apoptosis induced by CoCl2,cell vitality with significant difference compared with model group(P<0.05).Iridoids of V.jatamansi Jones can significantly improve apoptosis of PC 12 cells induced by CoCl2,which have potential value for prevention and treatment of neurodegenerative disease.
