Objective:To observe the effect of Scutellaria barbata on the epithelial-mesenchymal transition (EMT) of human colorectal adenocarcinoma epithelial cells SW480, and investigate its underlying mechanism.The aim is to elucidate the mechanism which inhibited EMT, further reveal the mechanism underpinning the anti-cancer invasion and metastasis of Scutellaria barbata and provide scientific experimental basis for clinical treatment of olorectal cancer.Methods:With Scutellarin and Luteolin as control product quality control of the medicinal materials fingerprint chromatography making Scutellaria barbata by high performance liquid.Following preparation of the ethanol extract of Scutellaria barbata, SW480 cells by cultivated in the different doses of Scutellaria barbata 1000?g/ml,500?g/ml,250 ?g/ml,125 ?g/ml,62.5?g/ml,31.25 ?g/ml in different time, the influence of Scutellaria barbata on the proliferation of SW480 cells was detected with MTT. Observed morphology of SW480 cells after treated with different dose of Scutellaria barbata by microscopy.The influence of Scutellaria barbata on the invasion of SW480 cells was detected with Transwell invasion assay.Using scratch test to detect migration of SW480 cells.The function of Scutellaria barbata on the transformation of SW480 cells was assessed by soft agar colony formation assay.The effects of Scutellaria barbata on the expression of miR-200a and the transcriptions of E-cadherin??-catenin?ZEB1?ZEB2?Vimentin involved in EMT were detected with quantitative real-time.Results:Scutellaria barbata at final concentrations of 125 ?g/ml,250 ?g/ml and 500 ?g/ml dose-dependently inhibited the proliferation,invasion and transformation of colonic cancer cells SW480 in vitro.Scutellaria barbata also suppressed the transcription expression of microRNA-200a,E-cadherin and down regulated the levels of Vimentin,?-catenin,ZEB1and ZEB2 mRNA in SW480 cells.Conclusion:Scutellaria barbata significantly inhibited cellular EMT-related events, including cell proliferation, migration, invasion and biotransformation, with the mechanism of regulating the expressions of microRNA-200a and its related genes.
