| Aging is an inevitable process of body functional decline,which increases the incidence of age-related diseases.Most anti-aging compounds are isolated from terrestrial plants,such as resveratrol and curcumin.In order to broaden the area to search for small molecular compounds against aging,researchers have turned attention to the ocean now.The diversity of marine natural products has great potential for discovering anti-aging compounds.In this study,yeast mutant strains K6001 was used as a bioassay system for screening samples from hundreds of marine natural products.The results showed that methanol crude extract of Ostrea edulis and Sagartiidae had anti-aging activity.Guided by the bioassay system,the EtOAc layer of Ostrea edulis was chromatographed on silica gel and ODS open column,and then purified by HPLC to obtain five active compounds(1~5).All of the five compounds could extend the replicative life of yeast(all of them are p<0.01).The chemical structures of thecompounds were determined as norcholesta-?5,? 22(E)?-? dien-?3β-? ol,24-methylene cholesterol,brassicasterol,crinosterol and cholesterol,by comparison of 1H NMR spectroscopic analysis and polarimetry with reported literatures.In addition,the EtOAc layer of Sagartiidae was separated by column chromatography and purified by HPLC to obtain a compound(6),guided by the bioassay system.It could also extend the replicative life of yeast(p<0.001).It was polyunsaturated fatty acid which the chemical structure was determined as 5,8,11,14,17-eicosapentaenoic acid,by comparison of 1H NMR and 13 C NMR spectroscopic analysis with reported spectroscopic data. |
