The Preventive Effect Of Faecalibacterium Prausnitzii Combined Kuikeling On Colitis In Rats And The Influence On Intestinal Microflora

Objective:To study the effects and the mechanism of Faecalibacterium prausnitzii (F. Prausnitzii) combined Kuikeling on colitis with rats with inflammatory bowel disease(IBD) and the influence on intestinal flora.Methods:Total of 50 rats was randomly divided into 5 groups, which named the control group, the model group, Kuikeling group, F.prausnitzii group and F.prausnitzii combined Kuikeling group, respectively. Each group was given medicine through administration by gavage for a week before model establishment. Colitis model was established by 2,4,6-trinitrobenzene sulfonic acid(TNBS)method. After 3 days, all rat were killed to observe the body weight and the colon length. Observing mucosal lesion area and colon mucosal inflammation and damage were assessed by the colorectal histological damage scores(HDS). Levels of interleukin-10 (IL-10), interleukin-12 (IL-12), tumor necrosis factor-a (TNF-?) and interferon-? (IFN-y)were detected by ELISA in the plasma, and then calculated the ratio of IL-10/IL-12. The expressions of IL-10 and TNF-a in the colon mucosal tissues was detected by immunohistochemical method. The rats faeces were quantified by RT-PCR to analyze amounts of F.prausnitzii, Bifidobacterium longum, Lactobacillus acidophilus, Escherichia coli in each group.Results:1.The symptoms of diarrhea were presented in all of the rats. Compared with the control group, body weight,colon length of rats had were significantly decreased, mucosal lesion area and the colon HDS were significantly increased (all P<0.001). Compared with the model group, the mucosal lesion area and the colon HDS were significantly decreased in Kuikeling group and F.prausnitzii combined kuikeling group (both P<0.05), F.prausnitzii combined kuikeling had reduced the body weight lost,the shorten of the colon (both P<0.05).Compared with the model group,the levels of IL-10 in the plasma and the ratio of IL-10/IL-12 were significantly increased (all P<0.05), F.prausnitzii combined kuikeling were very effective(P<0.001,P<0.003), the expression of TNF-a in the colon mucosal tissues were decreased (all P<0.05) and the level of TNF-a in the plasma was decreased, but this difference was not statistically significant in the preventive treatment groups.Compared with the model group, the levels of IL-10 the colon mucosal tissues were significantly increased and the level of IL-12 in the plasma were decreased in F.prausnitzii combined kuikeling group (both P<0.05), the level of IFN-? in the plasma was decreased statistically in Kuikeling group and F.prausnitzii combined Kuikeling group (P<0.001,P<0.05).Compared with the Kuikeling group, F.prausnitzii group,mucosal lesion area and the colon HDS were decreased, the levels of IL-12, IFN-y, TNF-a in the plasmaand the expression of TNF-a in the colon mucosal tissues were decreased, the levels of IL-10 in the plasma and the colon mucosal tissues,the ratio of IL-10/IL-12 were increased in F.prausnitzii combined kuikeling group, but this difference was not statistically significant.2.Compared with control group,content of F.prausnitzii and Lactobacillus acidophilus were decreased significantly (P<0.003,P<0.05), content of Escherichia coli were increased in the model group, F.prausnitzii groupJKuikeling group (P<0.003,P<0.01,P<0.01).Compared with the model group, the content of F.prausnitzii in F.prausnitzii group and F.prausnitzii combined Kuikeling group (P<0.05,P<0.003), the content of F.prausnitzii in Kuikeling group and the content of Bifidobacterium longum and Lactobacillus acidophilus in the preventive treatment groups were decreased,but there were no statistical significance. Compared with control group, B/E value of the model group,/\p rausnitzii group is decreased (P<0.001,P<0.05). Compared with the model group,B/E value of F.prausnitzii combined kuikeling group is increased (P<0.05). Compared with Kuikeling group,F.prausnitzii group,the content of F.prausnitzii,Bifidobacterium longum,Lactobacillus acidophilus and B/E value were increased, the content of Escherichia coli were decreased, but this difference was, not statistically significant.Conclusion:The preventive and therapeutic effects of F.prausnitzii combined Kuikeling is better than a single treatment and the mechanism may be related to the role of immunore gulation on inhibiting secretion of IL-12, TNF-a, IFN-y and promoting express of IL-10; F.prausnitzii combined Kuikeling can increase theintestinal probiotics, reduce pathogen, and had an effect on correcting intestinal dysbateriorsis.
