Objective:In this study the use of high frequency low amplitude vibration Power Plate Pro 5AIR versatile instrument training type of ovariectomized wistar rats of osteoporotic fracture intervention, preliminary study of mechanical vibration on rats of osteoporotic fracture healing rate and serum and affect bone metabolism related factors, and to further explore the mechanism of mechanical vibration fracture healing.Methods:1. After a week of adaptive feeding, the 45 3-month-old female Wistar rats were Conducted osteoporosis modeling, Intraperitoneal injection of 10% chloral hydrate(0.3ml / 100g) in rats,removal both ovaries of the rats through the back side bilateral incision into the abdominal cavity.2. fracture modeling: At the fifth month after Rat model of osteoporosis, 45 rats were anesthetized with chloral hydrate, cutting off the middle of the femur with a scalpel, and then using Kirschner wire(diameter 1mm) retrograde intramedullary fixation.3. vibration training intervention: the osteoporotic fracture rats were randomly divided into three groups: 15 rats were divided into sham ovariectomized group(Sham),15 rats were divided into ovariectomized group(OVX) and 15 rats were divided into ovarian vibration group(OVX-V). Vibration treatment: the Vibration group was placed on a vibration platform once a day for 15 min each time at 35 Hz,5 days a week. All groups of rats under anesthesia were shoot X-ray film to check the success of fracture modeling and fracture healing at the Second, fourth and sixth week after the fracture modeling surgery. The serum concentration of IL1- ?, IL-6, TNF-? and Tracp concentrations were measured by enzyme-linked immunosorbent assay(ELISA) at the Second, fourth and sixth week after the fracture modeling surgery.Results:1.Changes in rat fracture site callus structure:the fracture healing rate of the 3 groups sham-operated group(Sham), ovariectomized group(OVX) and ovariectomized vibration group(OVX-V) were compared to each other. X-ray examination showed that the healing rate of the vibration group was better than the control group or the sham group. The healing rate of the Sham group was better compared with the control group.2. Changes in blood biochemical2.1 Effect of mechanical vibration on ovariectomized rats in serum IL-1?((?)ąs)OVX-V group serum interleukin-1 ? concentration at 4 weeks, 6 weeks had a decreasing trend; serum interleukin-1? OVX group at 4 weeks, 6 weeks were increased trend; Sham group serum interleukin-1 ? concentration at at 4 weeks, 6 weeks had a decreasing trend; Different groups of serum concentrations of interleukin-1? at all the same time the overall comparison is OVX-V group <Sham group <OVX group. Sham group,OVX-V group compared with OVX group were significantly different.2.2 Mechanical vibration go on serum IL-6 in the ovary((?)ąs)The concentration of interleukin-6 in OVX-V group serum has a significantly reduced tendency at the fourth week and the sixth week; The concentration of interleukin-6 in OVX group serum has a increased tendency at the fourth week and the sixth week; The concentration of interleukin-6 in Sham group serum has a reduced tendency at the fourth week and the sixth week; The concentrations of interleukin-6 in Sham group, OVX-V group serum levels of interleukin-6 compared with OVX group at the same period in each individual, there are significant differences,except in the second week.2.3 Effect of mechanical vibration on ovariectomized rats serum of TNF-?((?)ąs)The concentration of TNF-?in OVX group serum has a reduced tendency at the fourth week and the sixth week; The concentration of TNF-?in Sham group serum has a reduced tendency at the fourth week and the sixth week;serum TNF-? Sham group at 4 weeks, 6weeks have tended to increase; The concentration of TNF-?in OVX-V group serum has a increased tendency in the fourth week and a has a reduced tendency in the sixth week;Different groups of serum concentrations of interleukin-1? at all the same time the overall comparison is OVX-V group <Sham group <OVX group, OVX-V group compared with OVX group were significantly different at the same time,there is no significant difference between Sham group compared with OVX group at the same time of all the periods.2.4 Mechanical vibration in ovariectomized rats serum Tracp((?)ąs)The concentration of Tracp in the 3 group serum all have a rising tendency at the fourth week but a tendency at the sixth week. Different groups of serum concentrations of interleukin-1? at all the same time the overall comparison is OVX-V group <Sham group<OVX group,The concentrations of tracp in Sham group, OVX-V group serum levels of interleukin-6 compared with OVX group at the same period in each individual, there were significant differences,except Sham group compared with OVX group in the fourth week.Conclusion:1.The situation of fracture healing of rats in each group were observed weekly by X-ray,follow-up X-ray showed the fracture healing rate of vibration group was better than thecontrol group. Mechanical Vibration can promote fracture healing of osteoporosis fractures of ovariectomized rats.2.vibration having a significant osteogenic effect, by promoting inflammatory cytokines interleukin-6 and interleukin-1? dissipation, inhibit the expression of tartrate-resistant acid phosphatase and the regulation of tumor necrosis factor to influence the generation of synthetic bone callus metabolism and the synthesis of bone metabolism, accelerate the process of osteoporotic fracture healing. Especially osteogenic mechanical vibration mechanism is unknown, but on the vibration after osteoporotic fracture related reports rarely,the paper results conclusions elaborate, hope can be more systematic, in-depth research and its clinical application for provide a theoretical basis.3.Preliminary studies indicate that the use of Power Plate Pro 5 AIR round training type instrument on rats is safe and effective, it is worth to promote the use of it and further study.
