The immune system is wrong to identify its own normal cells as antigen,cell immunity and/or humoral immunity,so that the body tissue damage,such a disease known as autoimmune diseases.The harm of autoimmune disease is self-evident,therefore,the study of the pathogenesis of autoimmune diseases for disease prevention,diagnosis and treatment has an important role.EAE is a widely used animal model for the study of multiple sclerosis.Polyunsaturated fatty acids can not be synthesized in the body,the main source is obtained from food.Polyunsaturated fatty acids play an important role in many functions of cell membranes and cells,so polyunsaturated fatty acids are essential for mammals.In previous studies,long chain n-3 polyunsaturated fatty acids play an important role in immune regulation.Long chain n-3 polyunsaturated fatty acids have been shown to attenuate the inflammatory response mediated by T cells in both human and animal models.DHA has a certain therapeutic effect on a variety of inflammatory diseases,such as atherosclerosis,asthma,arthritis.The specific molecular mechanism of DHA in inhibiting inflammation in multiple sclerosis is not particularly clear,and a thorough understanding of the pathological mechanisms of EAE disease contributes to the prevention and treatment of multiple sclerosis.After feeding the mice with DHA,we used MOG to model the EAE and observed the incidence of the disease.We found that the inflammatory response was reduced in mice fed DHA.The same results were obtained by detecting the secretion of inflammatory factors.Dendritic cells play an important role in the inflammatory response,such as initiation reaction,activation of T cells and phagocytosis.In the EAE model,we found that the ability of dendritic cells to activate T cells was different in the two groups.There were significant differences in the expression of costimulatory factors and cytokines in dendritic cells.In vitro differentiated dendritic cells also confirmed that DHA promotes the differentiation of dendritic cells.The DHA-treaded DCs stimulated by MOG,and their clinical manifestations were consistent with those of EAE mice fed with DHA.We can know that DHA can regulate the function of dendritic cells and enhance the immune tolerance.GPR120,also known as FFA4R,is a receptor for n-3 polyunsaturated fatty acids.Previous studies have found that GPRs is associated with inflammation.We detected the expression of GPR120 in different stages of differentiation of dendritic cells,and found that the expression of GPR120 protein was positively correlated with the maturation of dendritic cells.This suggests that GPR120 may play a role in the differentiation of dendritic cells.
