Objective: To observe the effect of acupoint injection on the Th17 /Treg balance of ossylin(OVA)in allergic rhinitis rats,and to explore the mechanism of acupoint injection in the treatment of allergic rhinitis.To provide experimental basis for clinical treatment of allergic rhinitis.Methods: 40 SD rats were randomly selected 8 to from normal group,the other rats were immunized with ovalbumin to reproduce the AR rat model.Successfully modeled rats were retained 24 and these rats were randomly divided into normal group,non-acupoint injection group and acupoint injection group,8 rats in each group.The general behavioral changes of the rats before and after treatment were observed and scored.Real-time RT-PCR was used to analyze Foxp3 mRNA expression in nasal mucosa.Immunohistochemistry was used to observe the expression of transcription factor 3(Foxp3)and retinoic acid-related orphan receptor(RORγt).The content of serum IL-17 was determined by ELISA.Results: 1.Behavioral symptom score: At the end of modeling,the behavioral scores of model group,non-acupoint injection group and acupoint injection group were more than 5 points,which was higher than that of normal group(P<0.05).These shows the success of modeling.Comparison between groups after treatment: After treatment,compared with the model group,the symptom score of the acupuncture group was lower(P<0.05),and the symptom score in the non-acupoint injection group were slightly lower,but the difference was not statistically significant(P>0.05);compared with non-acupoint group,the symptom score of the acupuncture group was decreased(P <0.05).Intra-group comparison after treatment : There was no significant difference in symptom score between normal group and model group(P>0.05);the symptom score of non-acupoint group was lower than that before treatment,but the difference was not statistically significant(P>0.05);the symptom score of the group was significantly lower than that before treatment(P<0.05).2.Real-time RT-PCR result: Compared with the normal group,the expression of Foxp3 m RNA in the nasal mucosa of the model group,non-acupoint injection group and acupoint injection group was decreased by real-time RT-PCR(P<0.05).Compared with the model group,the expression of Foxp3 in nasal mucosa of the acupoint injection group was increased(P<0.05),and there was no significant difference in the expression of Foxp3 mRNA in the nasal mucosa of the non-acupoint group(P>0.05);Compared with non-acupoint injection group,the expression of Foxp3 mRNA in nasal mucosa of acupoint injection group was higher(P<0.05).3.Immunohistochemistry results:Compared with the normal group,the expression of Foxp3 positive cells in nasal mucosa of the model group,non-acupoint injection group and the acupoint injection group was decreased(P<0.05);Compared with the model group,the expression of Foxp3 positive cells in nasalmucosa of the acupoint injection group was higher(P<0.05),and there was no significant difference between the non-acupoint injection group and the model group(P>0.05).Compared with the non-acupoint injection group,the expression of Foxp3 positive cells in nasal mucosa was higher(P<0.05).Compared with the normal group,the expression of RORγt positive cells in the nasal mucosa of the model group,non-acupoint injection group and acupoint injection group increased(P<0.05);compared with the model group,the expression of RORγt positive cells in nasal mucosa of acupoint injection group was decreased(P<0.05);non-acupoint injection group nasal mucosa RORγt expression positive cells showed no significant difference(P>0.05);compared with non-acupoint injection group,acupoint injection group nasal mucosa RORγt positive cells decreased significantly(P<0.05).4.Serum IL-17 content : Compared with the normal group,the serum IL-17 levels in the model group and non-acupoint group increased and the serum IL-17 level in the acupoint injection group was not statistically significant(P> 0.05).Compared with the model group,the serum IL-17 level in the acupoint injection group was significantly lower(P<0.05),and the serum IL-17 level in the non-acupoint injection group was not statistically significant(P>0.05);Compared with the non-acupoint injection group,the content of IL-17 in the acupoint injection group was significantly lower(P<0.05).5.The correlation between the detection indexes: There was a negative correlation between behavioral symptom score and Foxp3 mRNA expression in nasal mucosa and the expression of Foxp3 positive cells in nasal mucosa and there was a positive correlation between the expression of RORγt positive cells and serum IL-17 level in nasal mucosa.Conclusion : Acupoint injection can effectively alleviate the rats’ symptoms of allergic rhinitis,sneezing,runny nose and scratching nose,the mechanism may be through the up-regulation of Foxp3 expression to promote Treg cell formation,down-regulation of RORγt expression inhibition of Th17 cells differentiation and IL-17 production,regulation of allergic rhinitis rat Th17/ Treg cells balance. |
