Objective:To observe the different genotypes and different months of telomerase-deficient(Terc-/-)mice the characteristics of behavior change,and investigate acupuncture have expression influence on TRKB-ERK signaling pathways protein of the hippocampus region in telomerase-deficient mice brain.Methods:1.Choose the third generation of telomerase-deficient mice,and divide the mice when they grew up 3-4 weeks.Select Terc-/-and wild type mice after gene identification by PCR,and randomly selected for 2 months,3 months,5 months,7 months and 9 months of age mice every 8,a total of forty mice.Then the different months Terc-/-and wild type mice were made to behavior detection.After completion of testing,analysis of testing data,observation Terc-/-and wild type mice with changes in their behavior characteristics.2.Select twenty-four Terc-/-and wild type mice respectively,a total of forty-eight of them were divided into the control group,the acupuncture group and the electric acupuncture group.The control group was used in reared under the same conditions without any intervention.in the acupuncture group,acupuncture at the stomach 36(ST-36)position,once every five minutes twisting for 30 min/d for 7 d.In the electric acupuncture group,acupuncture at the stomach 36(ST-36)position,the eletroacupunture apparatus were connected with alternation of frequency and intensity of 2 mA,2/50 HZ,for 30 min/d for 7 d.Intervention take brain tissue next day after the treatment,for immuohistochemical staining and western blot methods on BDNF protein involved in signaling pathways expression in the hippocampus region.Results:1.Morris water maze experiment:Compared with the wild type mice,the homozygote mice have showed a trend of increase in its aging degree score and has a growth trend with the increase of months in the escape latency of sea trial,specially in 5,7,9 months it was statistically significant(P<0.05).2.the independent activity experiment:We obtained from the experimental results of the independent activities,there were no significant changes in the 2,3,5 months homozygote mice's autonomic activities within a fixed time compared with the same age of the wild type mice,but it's different in 7,9 months,the number of autonomic activities in the homozygote mice has apparently reduced and it have statistically significant(P<0.05).3.aging degree score:Compared with wild type mice,homozygous mice score values was no significant difference(p>0.05)when they are in 2 or 3 months,but the score values significantly higher when 5,7 and 9 months of age(p<0.05).4.Immunohistochemical:(1)the target surface density of the positive expressions of TRK-B protein in mice hippocampus region after treatment were:(homozygous)the control group(0.0233±0.007),the acupuncture group(0.0312±0.005),the electric acupuncture group(0.0363±0.007),compared with the control group,the electric acupuncture group was increased significantly(P<0.05).(2)the target surface density of the positive expressions of ERK protein in mice hippocampus region after treatment were:(homozygous)the control group(0.0148±0.074),the acupuncture group(0.0270±0.010),the electric acupuncture group(0.0201±0.008),compared with the control group,the acupuncture group was increased significantly(P<0.05).5.Western blot:(1)the relative volume of TRKB protein in mice hippocampus region after treatment were:(homozygous)the control group(0.6940±0.249),the acupuncture group(0.9525±0.139),the electric acupuncture group(0.9801±0.129),compared with the control group,the acupuncture group and the electric acupuncture group were increased significantly(P<0.05).(2)the relative volume of NF-kB protein in mice hippocampus region after treatment were:(homozygous)the control group(0.2891±0.070),the acupuncture group(0.4066±0.126),the electric acupuncture group(0.4856±0.066),compared with the control group,the acupuncture group and the electric acupuncture group were increased significantly(P<0.05).Conclusion:1.Compared with the same age wildtype telomerase knockout mice,the homozygote mice has apparently rheological change in their activities,and show a series of aging characteristics,particularly in 7,9 months.2.Acupuncture "ST-36" can promote the expression of TRKB?ERK and NF-kB proteins in homozygous mice hippocampus,but have no significant influence on the wild type mice.
