Background and Objective:Lung cancer in the global incidence of cancer in the first place,the number of deaths due to lung cancer each year about 1.4 million to 1.6 million,In recent years,the incidence of lung cancer and mortality are increasing rapidly.The development of lung cancer is affected by many factors,Smoking,ionizing radiation,lung chronic infection history and environmental pollution makes the incidence of lung cancer has become the first incidence of malignant tumors in China.The vast majority of lung cancer is originated in the bronchial mucosa epithelium,so lung cancer is also known as bronchial lung cancer,common lung cancer pathological type of non-small cell lung cancer and small cell lung cancer,Rather than small cell lung cancer,including squamous cell carcinoma,adenocarcinoma and large cell lung cancer,non-small cell lung cancer cases accounted for about 85%of all cases of lung cancer.Despite the continuous improvement in surgical procedures over the past few decades,radiotherapy,progression of chemotherapy,and the emergence of novel molecular-targeted drugs,the overall 5-year survival rate for lung cancer remains low at about 17.1%.The ubiquitin-proteasome system is the main cellular intracellular protein degradation pathway and plays an important role in many pathologies and physiological processes,including cell cycle regulation,apoptosis,inflammation,signal transduction and stress.This pathway is a reversible process in which Deubiquitrin(Dubs)is a protease that reverses the ubiquitination process and studies have shown that DUBs are involved in cell growth and apoptosis,hereditary tumors,neurodegenerative diseases,etc.Have played an important role.Ovarian tumor-associated protease(OTUD1)is one of the main members of the ubiquitination enzyme family.Ovarian tumor domain-containing protease 1(OTUD1)is widely expressed in many tissues of human,such as kidney,Intestinal,brain,liver,lung and so on.This study was to detect the expression of OTUD1 in non-small cell lung cancer(NSCLC)and its adjacent tissues by detecting the expression of OTUD1 in non-small cell lung cancer(NSCLC)and its clinical significance.Research methods:A total of 82 patients with non-small cell lung cancer were treated with thoracic surgery from Qilu Hospital of Shandong University from March 2008 to February 2009,At the same time forty patients with normal lung tissue were selected as negative control,The expression of OTUD1 in lung cancer tissues and adjacent tissues was detected by immunohistochemistry.Another from 2015 in 2015 to April 2015 in Shandong University Qilu Hospital thoracic surgery for primary non-small cell lung cancer in 25 cases,Real-time PCR was used to detect the expression of OUT1 in non-small cell lung cancer tissues and adjacent normal lung tissues.The clinical related pathological factors and follow-up data were analyzed in 82 patients.SPSS 19.0 statistical software was used to analyze the data,The rate of comparison between the two groups using ?2 test,Kaplan-Meier method and Log-rank test were used to analyze the difference of survival time,Cox regression multivariate analysis to determine the independent prognostic factors.Results:The expression rates of OTUD1 mRNA in non-small cell lung cancer tissues and adjacent tissues were 48.7%(40/82)and 12.5%(5/40)respectively.There was significant difference between the two groups(P<0.01).There was no significant correlation between OTUD1 expression and age(P = 0.386),sex(P = 0.392),smoking history(P = 0.632),pathologic type(P = 0.801),The expression of OTUD1 was significantly correlated with lymph node metastasis(P=0.005),differentiation(P=0.006)and TNM staging(P =0.015).Univariate and multivariate analysis showed that lymph node metastasis,TNM stage and OTUD1 expression were correlated with poor prognosis,P<0.01.Conclusion:OTUD1 is highly expressed in non-small cell lung cancer tissues and is closely related to the malignant progression and poor prognosis of non-small cell lung cancer.It can be used as a reference index to judge the biological characteristics and prognosis of non-small cell lung cancer.
