Object:The purpose of this paper is to investigate the therapeutic effect of aloin on acute trauma,further prepare graphene oxide hydrogel gel as a carrier of traditional Chinese medicine monomer aloe aloin of local drug delivery system,and discuss the effect and mechanism of aloin on wound healing and scar inhibition.Methods:1.A series of vitro and vivo experiments were used to investigate the therapeutic effect of aloin on skin wound.In vitro,the effects of aloin on cell proliferation and cell migration were investigated,and the effect of aloin on angiogenesis was studied by the tube formationexperiment.In vivo,the effect of aloin on wound healing rate was investigated by establishing a rat model of acute trauma.The woulnd skin was observed by scanning electron microscope(SEM),EGF(HE,Masson)and immunohistochemical staining(CD31 and EGF).2.The aloin-loaded graphene oxide gelwas constructed,and the prescription was optimized.Further,the morphological,stability and drug release were studied.Determination of Aloin in Gel by HPLC.3.The antibacterial activity in vitro of the aloin graphite oxide gel was investigated.In vivo experiments,we further investigated the skin irritation and wound healing of aloin oxidized graphene gel.By using SEM,HE and Masson trichrome staining,immunohistochemical staining of CD31 and ?-SMA,and testing the content of VEGF in the skin,the wound repair effect of the aloe gel was determinedResults:1.Aloin can spur on the proliferation of vascular endothelial cells and skin fibroblasts.The promoting effect of aloin on the migration ability of HSF cells was significant.Furthermore,aloin also had a certain effect on antibacterial,which demonstratespromise for promoting healing.The results of experiments in vivo showed that Aloin solution could improve wound healing rate.After treatment,the skin contains more new blood vessels with hair follicles and other appendages,and the collagen arrangement,similar to normal skin,was more regular and loose,which shows a certain anti scar effect.2.The content of each component in the prescription:carbomer:0.5%;graphene oxide:125?g/mL;aloin:lmg/mL triethanolamineadjust pH to 5.Aloin oxidized graphene gel exhibited fine consistency and moderate viscosity,a certain slow release function and good stability,which is suitable for topical skin administration.3.Aloin oxidized graphene gel was prepared according to prescription has good antibacterial activity.Skin irritation test also showed that it had no irritation to skin.In vivo,the results showed that the aloin oxidized graphene oxide gel could improve the wound healing rate.Through the investigation of the histological and immunohistochemical features of the healing skin,we found that the aloin oxidized graphene oxide gel could enchance the proliferation of new blood vessels.The healing skin contained a small amount of skin appendages which mainfests the new skin has a part of the normal function skin.Furthermore,the aloin oxidized graphene oxide gel showed a certain anti scarring activity.Conclusion:The aloin oxidized graphene oxide gel can be used as a good local drug delivery system to promote wound healing and reduce scar formation.
