Buckwheat contains many bioactive substances which allow it to have so many bioactivities such as antioxidant,antihypertension and antineoplastic.Most of the existing researches are focusing on the bioactivities of the exactraction from buchwheat.However,there is a lack of systematic quality evaluation of buckwheat,especially on the prevention of hypertension when we have buckwheat as food intake.Through this experiment,the hypotensive effect of buckwheat as food intake is proved and the mechanism is tested,in addition,a system is established to evaluate the level of buckwheat's hypotensive effect.Nine typical buckwheat were selected,and 110 male wistar rats were randomLy divided into 11 groups,which contain one normal control group?normal diet?,one model control group?8%NaCl+15%buckwheat?and nine experimental group?8%NaCl+15%buckwheat?.The artery systolic blood pressure was meatured on tail every week and the experiment lasted for 7 weeks.After that,the serum and the kidney tissue samples were collected.The level of hydroxyproline and the viability of ATPase was tested,the cellular morphology of kidney tissue was also observed under microscope.Some of the biochemical indexes including the level of Na+?Ca2+?NO?SOD?MDA in serum were tested.RT-PCR was used to measure the expressions of mRNA of Na+/K+-ATPase?PMCA1?ADD1?RRAR-?,and Western blot was used to test the expression of protein of Na+/K+-ATPase?Src?Ras?Raf?ERK?IKK?/??I?Ba??NF-?Bp65?PPAR-? in kidney tissue.The results showed that high salt diet would cause a significant rise in blood presure in wistar rats,and the 15%buchwheat diet can prevent the blood presure from rising,and reducing effcet of high salt diet on the Na+?Ca2+?NO?SOD?MDA level in serum and hydroxyproline level,viability of ATPase in kidney.It could also reduce the effect of high salt diet on the expression of mRNA of Na+/K+-ATPase?PMCA1?ADD1?RRAR-? in kidney tissue.Through increaseing the expression of Na+/K+-ATPase,buckwheat can regulate the expression of Src,Ras and Raf,and inhibit the expression of ERK,so that it can prevent the morphological change in kidney cells.Through regulating the expression of IKK?/? and I?B?buckwheat can inhibit the increace the expression of NF-?B p65 caused by high-salt diet,so that it can prevent the inflammation in kidney.The inflammation can also increase the expression of PPAR-y,and buckwheat can inhibit the inflammation by decrease the expression of NF-?B p65.These results all proved that the buckwheat diet can prevent hypertension that caused by high-salt diet.
