| Objective of experimental research:To establish model of type 2 diabetes by STZ and feeding high fat diet, thus wo can explore the Banxia Xiexin Decoction and its disassembled party’s mechanism of action that on concentration of blood glucose and serum insulin levels, insulin resistance index, liver glycogen, muscle glycogen and some synthesis enzyme that associated, so as to provide new therapeutic way of thinking to diabetes.Experimental method:Choosing SD rats,half male and half female as the experimental animals.Divided into normal control group (n=8) and for diabetic modeling group(total number is 112) randomly.The diabetic rats group were given high fat and high glucose diet for half a month,then injected the streptozotocin bacteria (STZ (45mg/kg) singlely to build the model of type 2 diabetes and the speed of injection is fast.Five days later,select the successful type 2 diabetic rats model that their fasting blood glucose between 11.1 mmol/L to 30.0mmol/L. According to the different levels of blood glucose, rats can be divided into diabetic model group,Banxia Xiexin decoction group, Xin Kai group, bitter Jiang group, Ganwei group. Diabetic model group were given normal saline irrigation, and the other groups were given corresponding drugs, the feeding time was 7 days. At the end of the experiment to detect the blood glucose of rats and experimental indexes.The experimental results:1.Banxia Xiexin decoction group, Ganwei group,bitter Jiang group can reduce the mortality of diabetic rats, to maintain the body weight of diabetic rats, but the Quanfang group’s effect is most obvious.2.Banxia Xiexin decoction group, bitter Jiang group, Ganwei group can lower blood sugar in diabetic rats, especially the Quanfang group hypoglycemic effect is obvious.3.Banxia Xiexin decoction group, bitter Jiang group, Ganwei group on the level of serum insulin in diabetic rats has certain promotion role, but it is not obvious, but to improve insulin resistance has obvious effect, Xinkai group were not regulation in aspects insulin secretion and insulin resistance.4. Ganwei group,Quanfang group can inhibit GSK-3 expression, improve the activity of GLUT4, promotethe the content of glycogen in liver and muscle tissue; bitter Jiang group and Xinkai group have no significant effect on GSK-3, can not effectively promote the expression of GLUT4 in the liver.Can’t promote the glycogen synthesis and metabolism effectively.5.Quanfang group, Ganwei group, bitter Jiang group, Xinkai group can improve the expression of MC4R that exist the diabetic model rat pituitary,so as to control the diet.Experimental conclusion:Banxia Xiexin decoction group and Ganwei group can significantly reduce the blood glucose and improve the insulin resistance and increase the glycogen content that in liver and muscle, its mechanism may have something to do with promoting the expression of GLUT4 in liver and inhibition of GSK-3 activity, reduce hepatic gluconeogenesis, enhance the body’s use of sugar; on the other hand may be associated with increased MC4R regulation of the satiety, reduced feeding on. And bitter Jiang group reduce blood glucose and improve the insulin resistance and promote the expression of GLUT4, but the relationship is not obvious, also can not inhibit GSK-3 activity effectively,thereby increasing transporter of glucose mediated, the hypoglycemic mechanism may be related to anti-inflammatory factors, need further experimental study; Xinkai Group alone is not obvious on early diabetic model rats, but will increase the mortality. |
