Objective To investigate the correlation between rs17038412 polymorphism of xanthine oxidase(XO)and high altitude Hyperuricemia(HUA)in male Han population.Methods A total of 186 male individuals with HUA and 54 male individuals without HUA(controls)of Han population were enrolled in this study,who moved to 4520 meters altitude of 0-10 years of life.We collected fasting elbow vein blood in the morning,determined serum uric acid and the hemoglobin content by the automatic biochemical analyzer,the mean pulmonary artery pressure by the B ultrasound,blood oxygen saturation,blood pressure,and the acquisition of heart rate and mean pulmonary artery pressure were collected at the same time.The concentration and activity were determined by ELISA method,genotypes of 10 polymorphisms were identified by polymerase chain reaction-high resolution melting analysis method.Results1.We detect serum uric acid concentration four times in three months,the incidence of HUA in the Han nationality of the plateau was 77.5%2.Hemoglobin(Hb)concentration in HUA group was significantly higher than those in control group,and blood oxygen saturation was significantly lower than those in control group(P < 0.01),while the age of NUA group,the exposure time,systolic blood pressure,diastolic blood pressure,heart rate and mean pulmonary arterial pressure(PAMP)were not significantly different from those in HUA group.3.Serum uric acid concentration and incidence of HUA were not different between different age groups(19 to 20 years,13 to 23 years old,>23 years old)and different altitude exposure time(0 to 12 months,13 to 36 months,>36 months)(P>0.05).4.The correlation analysis showed that there was a significant positive correlation between the concentration of XO and UA in the Han population of the Han nationality in the long term(P < 0.01),and the concentration of Hb increased with the increase of XO content in the plasma.The content of XO in HUA group was significantly higher than that in NUA group(P < 0.01).5.The frequencies of AT genotype of rs17038412 in HUA group(58.6%)was significantly higher than that in NUA group(37.1 %)(P = 0.05,OR = 3.96,95 %CI:1.42-11.08)Conclusion1.The incidence of hyperuricemia in male Han population in the plateau was as high as 77.5%.2.Early into the plateau,the occurrence rate of HUA can reach the highest peak within one year,and it does not change with the extension of the life time of the plateau.3.The concentration of uric acid in the initial period of entering the plateau can reach the peak,and it does not change with the extension of the life time of the plateau too.4.Plasma uric acid concentration was positively correlated with the concentration?activity of XO,and hemoglobin concentration,and negatively correlated with the saturation of oxygen.5.XO gene rs17038412 polymorphism of A allele may increase the risk of HUA in the Han nationality in the plateau,and the polymorphism of the AT gene is a risk factor for the occurrence of HUA in the Han nationality in the plateau.
