[Objective](1)To investigate the correlation between the expression of CEACAM1 expression in multiple myeloma and clinical characteristics and to evaluate the role of CEACAM1 as prognostic marker.(2)To investigate the role of CEACAM1 in the proliferation,cell cycle progression,apoptosis,migration and invasion of multiple myeloma cells.[Materials and methods](1)Between 2015.06 and 2015.10,50 cases of multiple myeloma patients were recruited in this study for sample collection.The CEACAM1 expression of samples was analyzed with flow cytometry and the correlation between CEACAM1 expression and clinical characteristics were analyzed.(2)A CEACAM1 overexpressing plasmid was constructed and transfected into U-266 and PRMI8266 cells to enforce CEACAM1 expression.The effect of CEACAM1 overexpression on cell proliferation,apoptosis,migration and invasion was determined by CCK-8 assay,flowcytometry,wound healing and Transwell assay,respectively.[Results](1)Among the 50 cases of patients,13 cases were Durie-Salmon stage I,19 cases were Durie-Salmon stage II and 18 cases were Durie-Salmon stage ?.(2)16 cases were CEACAM1 postive(CEACAM1 expression over 20%)while 34 cases were CEACAM1 negative(CEACAM1 expression less than 20%).CEAACM1 expression was psotively associated with CD33(r=0.467,P=0.038)with not significantlt associated with CD138 and CD56(P>0.05).(3)Overexpression of CEACAM1 in U-266 and PRMI8266 cells inhibits cell proliferation,blocks cell cycle progression,induces apoptosis,and suppresses migration and invasion.[Conclusion]The CEACAM1 expression on the surface of multiple myeloma is associated with the disease staging and prognosis,suggesting the potential of CEACAM1 as prognosis marker.Moreover,forced CEACAM1 overexpression in multiple myeloma cell line exerts anti-tumor effect,suggesting the role of CEACAM1 as tumor-suppressor gene.
