Objective: To observe the enhancement of anti-anoxia by Alpine Rhodiola oral liquid through animal experiment in vivo,while the mechanism of anti-anoxia was studied by cellular and molecular experiment.The objective of the present study was to provide the basis for rational use of Rhodiola preparation in clinical and social.Methods: Male ICR mice,male SD rats were served as experimental animal.Alpine Rhodiola oral liquid was given to rats and mice at the dose of 2ml/kg and 3ml/kg separately.The flunarizine hydrochloride suspension in which the concentration of flunarizine hydrochloride was 0.5mg/ml,chose as the positive control drug and it was prepared with flunarizine hydrochloride capsule and physiological saline.The suspension was administered to rats and mice at the dose of 1ml/kg and 1.5ml/kg separately.While sodium chloride injection was chose as solvent control drug.There were four parts in our animal experiment: the lethal experiments on normal pressure hypoxia rats,obturation hypoxia mice and hypoxia sodium nitrite poisoned mice,exhaustion swimming experiment on normal pressure hypoxia weight bearing mice.And the survival time of every rat or mice in these four parts was recorded.In the cellular and molecular experiment,the levels of lactate dehydrogenase,malondialdehyde,creatine kinase and superoxide dismutase of rat myocardial cells were measured under normal pressure hypoxia.The rat myocardial cells from SD neonatal rats were used to measure the cell apoptosis by flow cytometer following the requirements of cell apoptosis detection kit and cell activity was measured by MTT.The final data was shown by mean ± standard deviation(x? ± s).The data analysis was carried out by equal variance assumed t test between two groups.Results: Alpine Rhodiola oral liquid can significantly prolong the survival time of normal pressure hypoxia rats(p<0.01),obturation hypoxia mice(p<0.01),and hypoxia sodium nitrite poisoned mice(p<0.05),as well as the exhaustion swimming time of normal pressure hypoxia weight bearing mice(p<0.01).It can also significantly improve the activity(p < 0.01)and inhibit the of apoptosis(p < 0.01)of primary cultured rat myocardial cells.By Alpine Rhodiola oral liquid,the contant of SOD increased and the contant of LDH,CK and MDA decreases significantly.Conclusion: It is proved that under anoxic conditions,Alpine Rhodiola oral liquid has a significant anti-anoxia ability and it can also significantly improve the hypoxic exercise tolerance.Alpine Rhodiola oral liquid can obviously reduce the degree of lipid peroxidation induced by oxidative stress and protect cells from hypoxic myocardial injury,which is the possible mechanism of Alpine Rhodiola oral liquid's anti-anoxia effect.
