Objective: To observe the impact of different forms of transcutaneous acupoint electrical stimulation treatment after high-intensity exercise in rat cardiomyocytes autophagy, apoptosis,PKC? and PKC? expression situation.To study in rat cardiac response of high-intensity exercise and mechanisms of action of different forms of transcutaneous acupoint electrical stimulation of the heart protection, to provide a theoretical basis for transcutaneous acupoint electrical stimulation treatment in heart protection.Methods: Healthy male Sprague-Dauley(SD) rats 24, weight 190 ± 15 g, two months, randomly divided into four groups, each group 6,no weight-bearing exercise group(E), 6% loading exercise group(EG), 6% loading plus intermediate frequency electric acupoint stimulation preconditioning group(MEG), 6% loading plus intermediate frequency acupoint stimulation post-processing group(AEG), all of the group of rats exercise 2 hours. E and EG take heart immediately after 2 hours exercise; MEG should give a square wave of transcutaneous acupoint electrical stimulation for 30 minites before swimming half an hour, and then take heart immediately.AEG should give a square wave of transcutaneous acupoint electrical stimulation for 30 minites after swimming half an hour, and then take heart immediately. Stimulation of acupuncture points is Neiguan and Zusanli.By HE staining to observe cardiac morphology and microstructure, using Western blotting to detect myocardial cells' expression of Beclin1?LC3II/I, m TOR, PKC??PKC?; using tunel to detect the apoptosis of myocardial cells. Results: 1. One-time high-intensity exercise makes damage on morphology of rat myocardial cells. After transcutaneous acupoint electrical stimulation,MEG and AEG is better than EG on muscle fibers arrangement, mitochondrial morphology and nucleus shape, and MEG is a little better than AEG. The effect of transcutaneous acupoint electrical stimulation preconditioning is better than transcutaneous acupoint electrical stimulation post-processing. 2. To compared with E, apoptosis of EG were significantly increased(p < 0.01), the apoptosis level of MEG significantly reduced. Apoptosis of AEG also came down(p < 0.01), But the extent of reduction is less than MEG. It means one-time high-intensity exercise can induce apoptosis of cardiomyocytes, and transcutaneous acupoint electrical stimulation can lower apoptosis rate of myocardial tissue, transcutaneous acupoint electrical stimulation preconditioning better than transcutaneous acupoint electrical stimulation post-processing.3. The ratio of LC3II/I, EG have increased significantly more than E(p < 0.05), it prompted that high-intensity exercise can activate autophagy, the expression of MEG was higher than EG. It prompted that transcutaneous acupoint electrical stimulation preconditioning can further induce autophagy, and play a protective effect on the heart, MEG did not make significant change than AEG and EG. The expression of Beclin1, Compared with E, the expression of EG declined slightly, but the decline is very small and even can be ignored; but the expression of Beclin1 from MEG and AEG significantly increased(p <0.05), the expression level of AEG and MEG p value is not very different. MEG is slightly higher, but not significantly. To compared the expression m TOR with E, the expression of EG were significantly reduced(p < 0.01).The expression of AEG and MEG were significantly reduced to compared with E and EG(p < 0.01). AEG decreased more significantly than the MEG(p < 0.01). 4. The expression of PKC?, Compared with E,the expression of EG showed an upward trend, MEG and EG were occured downregulated phenomenon(p < 0.05), the gap between the expression of AEG and MEG is not obvious. One-time high-intensity exercise can increase the expression of PKC?, both transcutaneous acupoint electrical stimulation preconditioning and transcutaneous acupoint electrical stimulation post-processing can inhibit the over-expression of PKC myocardium after high-intensity exercise;and the effect is not very different.The expression of PKC? did not change significantly in each group. Each group are basically at the same level, there is no significant change. May PKC? not be sensitive to stimulate of One-time high-intensity exercise or transcutaneous acupoint electrical stimulation. It may be more sensitive response to cardiac hypertrophy or congestive heart failure caused by chronic fatigue movement. Conclusion: 1. One-time high-intensity exercise can both activate rat cardiomyocytes autophagy and apoptosis. 2. Transcutaneous acupoint electrical stimulation can increase cardiac autophagy, inhibit myocardial apoptosis, and plays a cardioprotective role.3. The effect is not obvious between transcutaneous acupoint electrical stimulation preconditioning and transcutaneous acupoint electrical stimulation post-processing in activating autophagy related proteins,and it is better in inhibition of apoptosis. 4. One-time high-intensity exercise increases the expression of PKC?,both transcutaneous acupoint electrical stimulation preconditioning and transcutaneous acupoint electrical stimulation post-processing can inhibit the overexpression of PKC? in rat cardiomyocytes, and play a role in myocardial protection.5. One-time high-intensity exercise and One-time transcutaneous acupoint electrical stimulation may no significant effect for PKC? expression in a short time.
