Object: To investigate the relationship between chemokine CXCR4 / SDF-1 in tongue squamous cell carcinoma and the metastasis of cervical lymph node,with clinical data analysis and vivo and vitro experiments to analysis the factors and mechanism of cervical lymph node metastasis.method: 1. Selecting tongue cancer patients a total of 165 cases from 2011 to December 2015 of Tianjin Stomatological Hospital. According to age, sex, the location of the primary tumor, tumor clinical staging, metastasis lymph node distribution of data packet analysis and arrangement, advanced tongue carcinoma and of squamous cell carcinoma of the tongue and the related influencing factors were statistically analyzed. Exploring tongue cancer and lymph node metastasis related factors.2. Taking 39 human tongue squamous cell carcinoma tissue checked of 2013 clinically, immunohistochemistry analysis the expression of CXCR4 and SDF-1.The discussion and analysis of the expression between CXCR4 / SDF-1 axis in tongue squamous cell carcinoma and lymph node metastasis.3. The application of Tca83 tongue squamous cell carcinoma cell line to establish an orthotopic transplantation tongue cancer model in nude mice.the mice were randomly divided into 3 groups, injecting Tca83 cells in suspension in the right lingual margin. Observe the growth of the tumor of tongue cancer animal model, and to investigate the expression of CXCR4 and SDF-1, the positive rate of CXCR4 and SDF-1. The statistical analysis SPSS17.0 software.Result: 1. 165 cases of tongue cancer patients overall lymph node detection transfer rate was 15.1 %, tongue cancer lymph node transfer mainly concentrated in the area I, II, III,but IV?V region were less transfered. 2.CXCR4 and SDF-1 were expressed in tongue carcinoma and lymph node tissue in different degree. And its expression is only related with lymph node metastasis,but not age, sex, tumor site, T stage or clinical stage.3.(1)Tca83 nude mice model was established successfully, and the tumor formation rate was 80 %. The tumor inhibition rates in low dose group(2)and high dose group were 38 % and 56 %, respectively. the greater the dose of AMD3100, the slower the growth of the tumor.(3)In 18 cases of tongue squamous cell carcinoma, 7 cases were detected positive expression, 38.9%. the cancer group was higher than that of normal tongue tissue, with statistical significance.Conclusion: 1. tongue cancer lymph node metastasis mainly in I, II, III District, IV District, V District less transfer. Lymph node metastasis was not related to age, sex, and location of lesions, but related to clinical stage and T stage. Only the clinical stage and cervical lymph node metastasis of tongue cancer related.2 CXCR4 expression is related with tongue cancer lymph node metastasis, age, sex, tumor site, but not T stage and clinical stage.3 Specific inhibitor AMD3100 of CXCR4 inhibits tumor growth in a dose-dependent, the inhibition gradually increased with the concentration. Through the inhibition of expression of CXCR4 and the metastasis of tumor lymph node, suggesting that the important role of SDF-1 / CXCR4 axis in the metastasis of lymph node of tongue cancer, and the SDF-1 / CXCR4 axis is one of the targets for anticancer therapy. But AMD3100 can not completely inhibit lymph node metastasis in squamous cell carcinoma of the tongue, and SDF1/CXCR4 axis is not the only factors of lymph node metastasis in squamous cell carcinoma of the tongue, other factors may also cooperate in regulating tumor metastasis.
