Objective:To investigate the effects of the Pu'er Tea Zhen and three main monomer components against inebriation(drunk) and hangover of alcohol using the drunk model in mice; to study the time-effect and dose-effect relationship of Pu'er Tea Zhen and three main monomer components so as to explain its action principle and provide the experimental data and scientific basis for the further exploitation.Methods:1. The mice were randomly divided into blank group and different dose experimental groups. The mice in each group were given distilled water and fed different doses of 56 degrees Red Star Erguotou. To observe and record the excited abnormal behavior(creeping instability, the hind legs dragged), loss of righting reflex(drunk), recovery time(sober) and the number of deaths of the mice, to determine the best drunk dose.2. Time-effect relationship: the mice were given Pu'er Tea Zhen(or distilled water) by ig. in each group, and 0, 5, 15, 30, 45 and 60 min later, 56 degrees Red Star Erguotou(or distilled water) were given by ig. To determine the blood concentrations of alcohol and acetaldehyde, the alcohol dehydrogenase and aldehyde dehydrogenase activity in stomach and liver, and AST/ALT; drew the time-effect curves of Pu'er Tea Zhen anti-inebriation and hangover.3. Dose-effect relationship: the mice were given different doses of Pu'er Tea Zhen(or distilled water) by ig. in each group, and 30 min later, 56 degrees Red Star Erguotou(or distilled water) were given by ig. To determine the blood concentrations of alcohol and acetaldehyde, the alcohol dehydrogenase and aldehyde dehydrogenase activity in stomach and liver, and AST/ALT; drew the dose-effect curves of Pu'er Tea Zhen anti- inebriation and hangover.4. The main ingredient hangover efficacy study: the mice were given Pu'er Tea Zhen and three main monomer components(or green tea or distilled water) by ig.in each group, and 30 min later, 56 degrees Red Star Erguotou(or distilled water)were given by ig. To determine the blood concentrations of alcohol and acetaldehyde,the alcohol dehydrogenase and aldehyde dehydrogenase activity in stomach and liver,and AST/ALT with Pu'er Tea Zhen and green tea for comparison.Results:1. All mice appeared drunk-action and no death after dose of 0.15 ml/10 g 56 degrees Red Star Erguotou by ig. All mice were awake three hours later.2. Compared with model group, the alcohol blood concentration of 30 min group at 30 min, 2 and 3 h decreased by 14.87%, 18.5%, and 31.0%, respectively,and acetaldehyde blood concentration decreased by 60.78%, 75.0% and 75.0%,respectively; the ADH activity in stomach and liver of 30 min group increased by0.24 and 0.38 times, respectively, and ALDH activity in stomach and liver of 30 min group increased by 0.42 and 0.37 times, respectively. Compared with blank group,the serum AST/ALT decreased significantly in the model group, while the experimental group decreased in some extend, ranging from 57.58%~66.67%; the tolerance time of 30 min group prolonged 1.21 times, and drunk sleep time shorten17.77%.3. Compared with model group, the alcohol blood concentration of M-experimental group at 2 h decreased by 34.12%, and acetaldehyde blood concentrations at 30 min, 2 and 3 h decreased by 65.52%, 89.29% and 86.89%,respectively; the ADH activity in liver of M-experimental group increased by 0.26 times, and ALDH activity in stomach and liver of M-experimental group increased by 0.31 and 0.43 times, respectively; the serum AST/ALT increased in the M-experimental group, ranging from 69.00%~76.09%; the tolerance time of M-experimental group prolonged 0.35 times, and drunk sleep time shorten 23.10%.4. Compared with model group, the alcohol blood concentration of Tea Polyphenols group at 2 h decreased by 23.27%, and acetaldehyde blood concentrations at 30 min, 2 and 3 h decreased by 76.19%, 89.04% and 86.44%,respectively. Compared with model and Pu'er Tea Zhen group, the ADH activity in liver of Tea Polyphenols group increased by 0.21 and 0.27 times, respectively, while the ADH activity in stomach was no significant difference. Compared with model and Tea Polyphenols group, ALDH activity in liver of Theabrownin group increasedby 0.23 and 0.17 times, respectively; the serum AST/ALT decreased in the experimental group, ranging from 53.19%~58.49%; the drunk sleep time of Tea Polyphenols shorten 27.13%.Conclusions:1. The dose of 0.15 ml/10 g 56 degrees Red Star Erguotou was the best for the drunk model.2. Time-effect relationship: the alcohol and acetaldehyde blood concentrations can significantly decreased after given Pu'er Tea Zhen by ig at 30 min, while the ADH and ALDH activities in stomach and liver were increased, so that reducing the liver damage; inhance the tolerance ability of mice for alcohol, and shorten the drunk sleep time.3. Dose-effect relationship: the alcohol and acetaldehyde blood concentrations can significantly decreased after given M-dose Pu'er Tea Zhen by ig, while the ADH and ALDH activities in stomach and liver were increased, so that reducing the liver damage; inhance the tolerance ability of mice for alcohol, and shorten the drunk sleep time.4. The main ingredient hangover efficacy study: the Tea Polyphenols can significantly decreased alcohol and acetaldehyde blood concentration and increased the ADH activity; the theabrownin can increased the ALDH activity in stomach and liver, the serum AST/ALT of three main monomer components were decreased, and the drunk sleep time of mice were shorten. Pu'er Tea Zhen has the anti-hangover ability similar to three components.
